Vindoline Inhibits RANKL-Induced Osteoclastogenesis and Prevents Ovariectomy-Induced Bone Loss in Mice

Abstract: Osteolytic bone diseases, for example postmenopausal osteoporosis, arise from the imbalances between osteoclasts and osteoblasts in the bone remodeling process, whereby osteoclastic bone resorption greatly exceeds osteoblastic bone formation resulting in severe bone loss and deterioration in bone structure and microarchitecture. Therefore, the identification of agents that can inhibit osteoclast formation and/or function for the treatment of osteolytic bone disease has been the focus of bone and orthopedic res… Show more

“…Bone quality maintenance is closely related with bone metabolism activity, mainly containing bone resorption and remodeling, which were driven by osteoclast and osteoblast activities ( Zhan et al., 2020 ). Therefore, osteoclast and osteoblast activities related enzymes and factors in serum were further investigated in this current research.…”

“…Osteoclasts and osteoblasts are key players in retaining bone mass and are the major cells involved in bone remodelling (Zhan et al, 2019). Several miRNAs that participate in the differentiation of osteoblasts have been identified.…”

“…During osteoclast differentiation, F-actin rings are considered as the characteristic structure of mature osteoclasts and is indispensable for its bone resorption activity. 23 In this study, we used Actin-Tracker to identify the formation and morphology of F-actin. Consistent with inhibition of osteoclast formation, the formation of podosomal actin belts was disrupted and the reduced actin ring formation was associated with intervention concentration of OB (Figure 3A).…”
