Villin1, a novel diagnostic marker for cervical adenocarcinoma

Abstract: The number of new cervical adenocarcinoma (AD) cases has risen slowly, however, its histological similarity to other tumor types and the difficulty of identifying the site of the original tumor makes the diagnosis of cervical AD particularly challenging. We investigated a novel molecular biomarker for cervical AD through the integration of multiple methods of genomic analysis. Tumor samples in discovery set were obtained from 87 patients who underwent radiotherapy, including 31 cervical AD. Microarray analysis… Show more

“…It is an anti-apoptotic epithelial protein that plays an essential role in regulating cellular morphology, survival and migration (26 were ECa and the remaining one case was diagnosed as small cell carcinoma of cervix, while no case of EMC was villin-positive (11). another study by Moll et al showed results similar to ours in that positive villin staining was found in only in 4/11 EMC (27).…”

“…Even if hysterectomy specimens were taken, it would be difficult to detect the original site of the carcinoma (10). Villin is an actin-binding protein which plays an important role in the maintenance of microvilli in epithelial cells and also has an essential role in the regulation of cell morphology and cell-specific epithelial anti-apoptotic mechanisms (11). Villin is also expressed in intestinal metaplasia and is associated with Barrett's esophagus and chronic atrophic gastritis, but it was not expressed in normal gastric and esophageal tissues (12).…”

“…We used some cervical cancer patients' tumor DNA containing HPV genome, which were detected in our previous experiments, as positive controls. The data indicated that the DNA samples used here showed good quality for PCR (24 Treatment. Patients were treated using a combination of external beam RT and high-dose rate brachytherapy.…”

“…Genomic DNA was also isolated from biopsies frozen in RNAlater (Ambion; Thermo Fisher Scientific, Inc., Waltham, MA, USA) using the Genomic-tip 100/G kit (Qiagen GmbH). HPV genotypes were determined using the polymerase chain reaction (PCR) method (22) and the Linear Array HPV Genotyping test according to the manufacturer's instruction (Roche Molecular Diagnostics, Pleasanton, CA, USA) (23,24). Samples that contained more than about 70% of tumor cells were used in our study (22).…”

“…In this issue of Cancer Biology & Therapy, Nakamura and colleagues investigate a novel molecular biomarker for cervical AD through genomic analysis. 4 Nakamura et al focused on chromosomal regions where genetic alterations are frequently observed in cervical cancer. Chromosome 2q33.3-q36 was reported as a frequently deleted locus by array-CGH.…”
