Video-rate multi-color structured illumination microscopy with simultaneous real-time reconstruction

Abstract: Super-resolved structured illumination microscopy (SR-SIM) is among the fastest fluorescence microscopy techniques capable of surpassing the optical diffraction limit. Current custom-build instruments are able to deliver two-fold resolution enhancement with high acquisition speed. SR-SIM is usually a two-step process, with raw-data acquisition and subsequent, time-consuming post-processing for image reconstruction. In contrast, wide-field and (multi-spot) confocal techniques produce high-resolution images inst… Show more

“…The laser is coupled into a high-power single-mode fiber to clean up its beam profile and to enable its easier adjustment of the illumination blaze angle with respect to the DMD surface. The grating patterns displayed on the DMD are calculated via a (modified) search algorithm used to find suitable binary gratings [9,11,27]. The light diffracted off of the DMD is subsequently collected by a lens and the polarization of the excitation beam is set by a segmented polarization filter ( fig.…”

“…Lastly, the industry-grade camera is used to detect the fluorescence signal with tube lens L7 which results in a projected pixel size of 70 nm and 35 nm for SIM (more details in appendix A.1 and appendix A.2). The raw image data is fed into a GPU-based real-time data processing system [11]. A detailed description of (a) and (b) can be found in appendix A.1.…”
