This review describes biofilms and the need for a materials science/engineering approach to solve the problems in the medical field. In particular, biofilm problems are closely related to infectious diseases. Most chronic diseases and the hospital-acquired infections could be attributed to biofilms. Biofilms usually form on materials. They may be biomaterials such as implants, catheters, stents and others. These films can also form on items outside of the human body. They include beds, service tables, medical knives, needles for injections, etc. Even though materials obviously affect biofilms formation and growth, there have been few studies using the materials science approach. In this review, we summarize the concept of biofilms from the viewpoint of materials science. Topics include the interaction between biofilms and materials (especially metallic materials), evaluation techniques, a bird's-eye analysis of previous investigations, and a discussion about the future direction for developing anti-infectious metallic materials.

Biofilms, mainly comprised of bacteria, form on materials’ surfaces due to bacterial activity. They are generally composed of water, extracellular polymeric substances (polysaccharides, proteins, nucleic acids, and lipids), and bacteria. Some bacteria that form biofilms cause periodontal disease, corrosion of the metal materials that make up drains, and slippage. Inside of a biofilm is an environment conducive to the growth and propagation of bacteria. Problems with biofilms include the inability of disinfectants and antibiotics to act on them. Therefore, we have investigated the potential application of alternating electromagnetic fields for biofilm control. We obtained exciting results using various materials’ specimens and frequency conditions. Through these studies, we gradually understood that the combination of the type of bacteria, the kind of material, and the application of an electromagnetic field with various low frequencies (4 kHz–12 kHz) changes the circumstances of the onset of the biofilm suppression effect. In this study, relatively high frequencies (20 and 30 kHz) were applied to biofilms caused by Escherichia coli (E. coli) and Staphylococcus epidermidis (S. epidermidis), and quantitative evaluation was performed using staining methods. The sample surfaces were analyzed by Raman spectroscopy using a Laser Raman spectrometer to confirm the presence of biofilms on the surface.

Effects of Ionic Liquids on Biofilm Formation in a Loop-Type Laboratory Biofilm Reactor

Biofilm formation was carried out using low concentrations of ionic liquid (IL) to investigate the potential control of biofilm formation with IL. A 2000ml reservoir was filled with tap water with/without a low concentration of ionic liquid in aqueous solution. The apparatus (Laboratory Biofilm Reactor: LBR) was a loop type Laboratory Biofilm Reactor (LBR) composed of an acrylic pump and a bottom tank. A pump circulated the water in the loop system of the laboratory biofilm reactor (LBR). The glass specimens were immersed in the LBR with/without a low concentration of ionic liquid in aqueous solution for 2 or 7days. They were observed and evaluated by using Crystal Violet, a 3D optical microscope and a Raman spectrometer to determine their biofilm behavior. The results were discussed from the viewpoint of a cooling pipe system simulation. We found out and confirmed that the formation of biofilm was controlled when a low concentration of aqueous solution of ionic liquids was circulated.
