In vivo Action of Corticosteroids on the Guinea Pig Lens

The influence of intravitreal injection of corticosteroids in guinea pigs on lens cation concentration, lens density and leucine-l-[¹⁴C] incorporation into the lens protein was examined. 6-methylprednisolone acetate (Urbason-Kristallsuspension) led to a temporary decrease of lens density and a corresponding sodium increase. Leucine incorporation was inhibited for at least 2 weeks. Aldosterone and different solvents did not influence the leucine incorporation significantly. The presence of subcapsular vacuoles, observed in about 50% of the methylprednisolone-treated lenses, coincided with the raised sodium levels. However, the authors believe that the inhibition of the protein synthesis is more important with respect to steroid-cataract development than the transient sodium influx.
