Venezuelan Equine Encephalitis Virus nsP3 Phosphorylation Can Be Mediated by IKKβ Kinase Activity and Abrogation of Phosphorylation Inhibits Negative-Strand Synthesis

Abstract: Venezuelan equine encephalitis virus (VEEV), a mosquito transmitted alphavirus of the Togaviridae family, can cause a highly inflammatory and encephalitic disease upon infection. Although a category B select agent, no FDA-approved vaccines or therapeutics against VEEV currently exist. We previously demonstrated NF-κB activation and macromolecular reorganization of the IKK complex upon VEEV infection in vitro, with IKKβ inhibition reducing viral replication. Mass spectrometry and confocal microscopy revealed an… Show more

“…and 1146–1164 aa. in other regions) were prepared as B cell epitopes, which partially overlapped with previous reports 21 26 . In this study, antibodies against the RBD were not evident, but only a few antibodies recognized these B cell epitopes in the RBD with high neutralizing activity.…”

“…Antibodies targeting the RBD and S protein have enhanced potential for providing cross-protective immunity 21 . The bioinformatics approach has been rapidly reported to identify potential B and T cell epitopes in S protein, which has provided data regarding antigen presentation, antibody-binding properties, predicted evolution of epitopes 21 26 , and interaction with immune sensors 27 29 . A list of glycoprotein amino acid positions having a high probability of predicted B cell epitopes has been compiled.…”

“…The generated helper T cells (Th1, Th2, and Th17) present the viral antigens to the B cells (differentiated plasma cells), which produce CoV-specific antibodies or Ig required to neutralise the virus (Fig. 3 ) (Li et al 2017 , 2020a ; Ahmed et al 2020 ; Huang et al 2020 ). Several studies have shown association between higher antibody production with stronger helper T cell cytokine secretion in SARS-CoV infection (Peng et al 2006 ; Li et al 2008a ; Fan et al 2009 ).…”

“…Research on COVID-19 prevention through this approach was begun immediately after release of relevant basic data such as its full genomic sequence. Specific epitope regions in SARS-CoV-2 with high homology to SARS-CoV, the best-characterized coronavirus in terms of epitope responses, were identified 17 19 . Antigenic properties of spike glycoprotein were more focused by theory and experimental researchers 20 22 .…”
