VEGF Expression in Hypoxia and Hyperglycemia

Abstract: ABSTRACT. Vascular endothelial growth factor (VEGF), an angiogenic factor for endothelial cells, is produced by glomerular and tubular epithelia. Using immunoelectron microscopy, VEGF expression by podocytes (GEC) and the proximal tubular epithelium of rat kidney was confirmed. To elucidate the mechanisms of VEGF production and its physiologic consequences, studies were performed in cultured GEC and proximal tubular epithelial cells (RPTEC). Both GEC and RPTEC expressed VEGF-120 and 164 mRNA, as detected by qu… Show more

“…PCR was performed in a DNA Thermal Cycler with the primers mentioned below. Oligonucleotide primers were designed to amplify VEGF mRNA and identify unique VEGF isoforms [24] . The sequence of the sense primer was 5'-GGACATCTTCCAGGAGTACC-3', and the sequence of the antisense primer was 5'-GTTC-CCGAAACCCTGAGG-3' .…”

“…Of the cytokines identified in the ADPKD liver cyst fluid, VEGF, monocyte chemoattractant protein, angiogenin, IL-6, and IL-8 can stimulate angiogenesi~.~3-~~ Hypoxia is a potent stimulant for production and secretion of angiogenic factors. [27][28][29] Consequently, EM was used to evaluate whether the morphological features of cystic epithelial cells mirror the features of hypoxic cholangiocytes. Superficial cyst walls from three different ADPKD patients each shared common histological features.…”

“…2 Often associated with DPM is an abnormal ramification pattern of the portal vasculature in close vicinity to cysts and microhamartomas ("pollard willow" pattern). 3 Endothelial and epithelial cells are known to exchange multiple paracrine signals, 4 particularly during morphogenesis and tissue remodeling. 5,6 A variety of different signals and, in particular, angiogenic growth factors and their receptors is involved in the regulation of the vascular growth and differentiation.…”
