An efficient plant regeneration protocol for shoot organogenesis from Hovenia dulcis callus cultures was established. Induction of organogenic callus was achieved on Murashige and Skoog (MS) medium supplemented with 4.65 lM kinetin and 4.5 lM 2,4-dichlorophenoxyacetic acid (2,4-D). Further differentiation of organogenic callus into primordia, shoot-like structures, and plantlets was achieved on MS medium supplemented with 0.23 lM gibberellic acid (GA 3 ) and 0.46 lM kinetin. Numerous abnormal shoots developed upon transfer of callus to MS medium containing cytokinins, and these failed to grow further into whole plantlets. However, transfer of 'abnormal' shoots to a fresh MS medium lacking cytokinins resulted in growth of normal shoots. Elongated shoots subsequently were rooted in basal MS medium, and whole plantlets were established in a soil mix. Analysis of regenerated plants using random amplified polymorphic DNA (RAPD) confirmed the genetic stability of these regenerant plantlets.

Despite that there is some literature on pollen morphology of Rhamnaceae, studies addressing general aspects of the microsporogenesis, microgametogenesis, and anther development are rare. The aim of this paper is to describe the ultrastructure of pollen grain ontogeny with special attention to tapetum cytology in Hovenia dulcis. Anthers at different stages of development were processed for transmission and scanning electron microscopy, bright-field microscopy, and fluorescence microscopy. Different histochemical reactions were carried out. The ultrastructural changes observed during the development of the tapetal cells and pollen grains are described. Large vesicles containing carbohydrates occur in the tapetal cell cytoplasm during the early stages of pollen development. Its origin and composition are described and discussed. This is the first report on the ontogeny and ultrastructure of the pollen grain and related sporophytic structures of H. dulcis.

Hovenia dulcis T. conhecida como uva-do-japão é uma planta originária do Himalaia, possui frutos saborosos e ricos em açúcares quando maduros. É difundida no sul e no sudeste do Brasil. Há poucos trabalhos científicos explorando tal espécie e não há relatos de seu aproveitamento em produtos alimentícios ou bebidas. Esta pesquisa teve como objetivo a determinação da sua composição centesimal e a produção de fermentado de uva-do-japão. A fruta apresentou elevada quantidade de açúcares (açúcar redutor em glicose e açúcar não redutor em sacarose de 11 e 11,8% (m m-1), respectivamente, fibras bruta (2,37% m m-1), e baixo conteúdo de lipídeos (0,10% m m-1). A polpa obtida dos frutos foi submetida à fermentação alcoólica e originou fermentado com teor alcoólico de 7,2 ºGL, com potencial para produção de vinagre. O fermentado de uva-do-japão tinha coloração amarela-acastanhada, levemente turvo, com odor agradável e adocicado, característico da fruta. Com acidez de 0,82% (g 100 g-1) e pH de 4,01, apesar do pH ácido, o produto apresentou residual de açúcares de 3,44% (g 100 g-1), sabor relativamente doce e agradável, combinando com a sua acidez. Os parâmetros analisados na fermentação apresentaram produção final de etanol de 56,8 g L-1 e rendimento de 0,51 g L-1. A eficiência da fermentação alcoólica foi de 99,8%. O processo fermentativo findou após 96 horas e a produtividade alcançada foi de 0,59 g L-1 h-1. Palavras-chave: Fermentado de fruta. Hovenia dulcis T. Uva-do-japão. Vinho.
