VCAM-1 Expression on Bone Marrow Stromal Cells from Patients with Myelodysplastic Syndromes

Abstract: We studied the expression of VCAM-1 adhesion molecules on stromal cells from the bone marrow of patients with myelodysplastic syndromes, healthy donors, and patients with chronic myeloproliferative diseases and acute leukemias. Expression of adhesion molecule on mesenchymal stromal cells from the bone marrow of patients and healthy donors was evaluated after 2-4 passages by the methods of immunoprecipitation and electrophoresis. VCAM-1 expression in the majority of patients with myelodysplastic syndromes was l… Show more

“…VLA‐4 expression has been associated with chemoresistance in AML. It is an integrin that can bind to VCAM‐1, which is expressed by endothelial and stromal cells, as well as fibronectin (Ulyanova et al , ; Lubkova et al , ). In contrast to endothelial cells, only low levels of ICAM‐1 or VCAM‐1 are expressed on stromal cells (Peled et al , ).…”

“…CXCR4 activation increases the affinity between VCAM1 expressed on the surface of MSC and its receptor VLA4, expressed by HSC and MK. Both pathways, CXCL12/CXCR4 and VCAM1/VLA4 are implicated with HSC migration (Lubkova et al . 2011; Moll and Ransohoff.…”

“…MSCs secrete the chemokine CXCL12 and Vascular Cell Adhesion molecule-1 (VCAM1) which bind to their respective receptors, CXCR4 and VLA4, expressed upon the surface of HSCs. (Lubkova et al . 2011; Liekens et al .…”

“…The establishment of HSPCs in hematopoietic locations is mediated by the expression of CXCL12 and vascular cell adhesion molecule-1 (VCAM-1), which bind to CXCR4 and to very late antigen-4 (VLA-4), respectively (29,(38)(39)(40)(41). It is well known that, after infection, in both classical BM and EMH hematopoiesis, HSPCs are activated to produce mature lineage cells to fight pathogens.…”
