The morphology and composition of the three otoliths of the Antarctic ice-fish Chionodraco hamatus were studied by scanning electron microscopy and X-ray diffraction. The composition of the sagitta, lapillus and asteriscus protein matrices was also analysed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis, western blots and confocal laser scanning microscopy to reveal the presence of and to localize the calcium-binding proteins calmodulin, calbindin and S-100. Morphological results indicated that the otoliths in this ice-fish were similar to those of Trematomus bernacchii , a red-blooded Antarctic species [B. Avallone et al. (2003) J. Submicrosc. Cytol. Pathol. 35 , 69-76], but rather different from those of other teleosts. These two Antarctic species possessed a completely vateritic asteriscus, whereas their sagitta and lapillus were made mostly of aragonite. Parallel analysis of protein patterns in C. hamatus and T. bernacchii revealed that the sagitta significantly differed from the lapillus and asteriscus in both species. The sagitta did not contain the S-100 protein and showed calmodulin and calbindin located in discontinuous or incremental zones, respectively. These results demonstrate that the otoliths of C. hamatus and T. bernacchii share more resemblances than differences and support the idea of a common origin of these species.

Otoliths, calcium carbonate (CaCO 3 ) ear bones, are among the most commonly used age and growth structures of fishes. Most fish otoliths are comprised of the most dense CaCO 3 polymorph, aragonite. Sturgeon otoliths, in contrast, have been characterized as the rare and structurally enigmatic polymorph, vaterite-a metastable polymorph of CaCO 3 . Vaterite is an important material ranging from biomedical to personal care applications although its crystal structure is highly debated. We characterized the structure of Lake Sturgeon otoliths using thermal analysis and neutron powder diffraction, which is used non-destructively. We confirmed that while Lake Sturgeon otoliths are primarily composed of vaterite, they also contain the denser CaCO 3 polymorph, calcite. For the vaterite fraction, neutron diffraction data provide enhanced discrimination of the carbonate group compared to x-ray diffraction data, owing to the different relative neutron scattering lengths, and thus offer the opportunity to uniquely test the more than one dozen crystal structural models that have been proposed for vaterite. Of those, space group P6 5 22 model, a = 7.1443(4)Å, c = 25.350(4)Å, V = 1121.5(2) Å 3 provides the best fit to the neutron powder diffraction data, and allows for a structure refinement using rigid carbonate groups.Vaterite, a metastable polymorph of calcium carbonate (CaCO 3 ) 1 is of substantial interest as a naturally occurring biomaterial, and for its use as an additive in various consumer products ranging from paper and coatings to plastic and elastomer reinforcement to food supplements, oral hygiene aids, and cosmetics 2 . For instance, vaterite's greater ability to dissolve in body fluids compared to other polymorphs of CaCO 3 makes it desirable for manufacturing nanocapsules for delivering drugs at the cellular level 3 . Despite the realized and potential importance of vaterite to humans, some of the most basic facts about this compound such as its crystalline structure have challenged researchers. Currently, more than a dozen crystal structure models have been proposed with little weight favoring one over the others 1 . Although most otoliths of teleost fishes contain aragonite, a detailed survey of the otoliths of serveral species confirms that other crystalline forms of calcium carbonate occur. Otoliths of Hoplostethus atlanticus, Pagrus major, Macruronus novaezelandiae, Merluccius australis, Congiopodus coriaceus, Kathetostoma giganteum, Argentina elongata, Rhomobosolea tapirina, Neophrynichthys latus, Coelorinchus aspercephalus, Paranothothenia microlepidota, and Gonorhynchus gonorhynchus contained the aragonite, calcite, and vaterite morphs of calcium carbonate in varying proportions. Aragonitic otoliths of Allocyttus niger, Hoplostethus altlanticus, and Pagrus major contained sequences of calcite-like crystals. The surface of the vateritic otolith of Acipenser brevirostrum is shown in detail. Three classes of information are stored in the crystalline structure of the otolith: shape conservation, coexisting crystal morphs, and consecutive changes in crystal morph. Analysis of this crystalline information supports the hypothesis of control of growth of the otolith by proteins from the sensory epithelium or macula. Protein variation involved may be genetic in origin, or non-genetic arising from "stuttering" of the translation process. Proteins extracted from vateric and aragonitic morphs of the otolith of Macruronus novaezelandiae showed differences in infared absorption spectra that were consistent with two different amino acid sequences. © 1993 Wiley-Liss, Inc.
