Vascular Growth Factors and Glomerular Disease

Abstract: The glomerulus is a highly specialized microvascular bed that filters blood to form primary urinary filtrate. It contains four cell types: fenestrated endothelial cells, specialized vascular support cells termed podocytes, perivascular mesangial cells, and parietal epithelial cells. Glomerular cell-cell communication is critical for the development and maintenance of the glomerular filtration barrier. VEGF, ANGPT, EGF, SEMA3A, TGF-β, and CXCL12 signal in paracrine fashions between the podocytes, endothelium, a… Show more

“…Foot processes use an integrin-based protein complex to attach and deliver key components to the underlying glomerular basement membrane (GBM) and thereby to communicate with endothelial and mesangial cells [38, 39]. Maintaining these critical foot process inter-relationships under hypertrophying conditions, particularly when rapidly occurring over short periods of time, must impose remarkable logistical challenges in space and time that have to be accommodated by as yet poorly understood signaling mechanisms.…”

“…Correct assembly of the glomerular vasculature is required for filtration barrier function (Bartlett et al, 2016). At p17, Tfeb iECÀ/À kidneys were smaller (% area of Tfeb iECÀ/À mice versus control 72.6 AE 7.9%; mice n = 3, P < 0.01), characterized by poorly developed glomeruli ( Fig EV3B) and reduced volumes (20.53 AE 2.50 × 10 4 lm 3 for control mice and 14.43 AE 3.27 × 10 4 lm 3 for Tfeb iECÀ/À ; mice n = 3, P < 0.002).…”

“…Diabetic vascular dysfunction also affects various other organs and tissues in extremities, heart, nerves, wounds and kidneys. The contribution of the Ang–Tie system to the pathogenesis of ocular and diabetes-associated vascular diseases is a field of intense research [117,120]. …”
