Varying modalities of perinatal exposure to a pesticide cocktail elicit neurological adaptations in mice and zebrafish

Abstract: Highlights 1) Perinatal pesticides links with long-term behavioral modifications in male mice.2) Behavioral changes are accompanied by electrographic brain slowing in vivo.3) In zebrafish larvae, pesticide cocktail elicited dose-dependent motor-behavioral changes.4) In zebrafish larvae, chlorpyriphos is a behavior-modifying pesticide contained in the cocktail.5) Non-lethal prenatal pesticide cocktail does not provoke brain malformations.

“…The current findings revealed substantial overexpression of the GFAP gene in the TMX-intoxicated group. These findings are consistent with previous studies that found elevated GFAP expression to be a sensitive marker of neurotoxins such as imidacloprid and thiacloprid (Katić et al 2021 ; Forner-Piquer et al 2021 ; Abomosallam et al 2023 ). These changes may indicate higher dopaminergic and serotonergic turnover in the brain (Abd-Elhakim et al 2018 ).…”

“…The total of animals used as controls was 1112, whereas the exposure individuals counted 2045. The highest concentration of fungicide in the meta-analysis was 20 mg/L for triadimefon 63 , while the lowest was 0.0001 mg/L for mepanipyrim, ziram, tiophanate, captan, and boscalid 26 , 64 .…”

“…The Y-maze apparatus had three identical arms (40 × 9 × 16 cm) placed at 120° with respect to each other. As previously described (Forner-Piquer et al, 2021), each mouse was placed at the end of one arm and allowed to explore freely the apparatus for 5 min. Spontaneous alternation performance (SAP) was assessed by scoring the pattern of entries into each arm during the 5 min of the test.…”
