Greater availability of leaf dark respiration (Rdark) data could facilitate breeding efforts to raise crop yield and improve global carbon cycle modelling. However, the availability of Rdark data is limited because it is cumbersome, time consuming, or destructive to measure. We report a non‐destructive and high‐throughput method of estimating Rdark from leaf hyperspectral reflectance data that was derived from leaf Rdark measured by a destructive high‐throughput oxygen consumption technique. We generated a large dataset of leaf Rdark for wheat (1380 samples) from 90 genotypes, multiple growth stages, and growth conditions to generate models for Rdark. Leaf Rdark (per unit leaf area, fresh mass, dry mass or nitrogen, N) varied 7‐ to 15‐fold among individual plants, whereas traits known to scale with Rdark, leaf N, and leaf mass per area (LMA) only varied twofold to fivefold. Our models predicted leaf Rdark, N, and LMA with r2 values of 0.50–0.63, 0.91, and 0.75, respectively, and relative bias of 17–18% for Rdark and 7–12% for N and LMA. Our results suggest that hyperspectral model prediction of wheat leaf Rdark is largely independent of leaf N and LMA. Potential drivers of hyperspectral signatures of Rdark are discussed.

SummaryLeaf optical properties can play an important role in determining the red/far-red light ratio, a signal of impending competition, in plant canopies. Knowledge of leaf optical properties and factors affecting them is important in understanding of the impacts of red/farred ratio in agroecosystems. Effects of leaf position on the plant stem on their optical properties at 660 and 730 nm were studied in tomato and two weeds Chenopodium album and Amaranthus retroflexus. Leaf position on stem strongly influenced leaf optical properties. Reflectance and transmittance were generally lower for the C. album and A. retroflexus leaves at higher positions on the stem, except for reflectance at 730 nm in C. album, which did not change. Reflectance was not affected in tomato. Transmittance generally decreased for leaves at higher positions. Red/far-red ratios of reflected (R ratio ) and transmitted (T ratio ) light generally decreased in all species, except R ratio in tomato, where it increased slightly at higher positions. These effects were greater in A. retroflexus compared with C. album and tomato. Changes in these ratios were partly explained by chlorophyll content and leaf mass per area. The results show that leaf position on plant stem influences leaf optical properties in tomato and two weeds and this effect differed between species. These influences and the differences among species could modify red/far-red ratios in canopies comprising these species, which could influence their growth and inter-plant interactions.

Leaf area growth determines the light interception capacity of a crop and is often used as a surrogate for plant growth in high-throughput phenotyping systems. The relationship between leaf area growth and growth in terms of mass will depend on how carbon is partitioned among new leaf area, leaf mass, root mass, reproduction, and respiration. A model of leaf area growth in terms of photosynthetic rate and carbon partitioning to different plant organs was developed and tested with Arabidopsis thaliana L. Heynh. ecotype Columbia (Col-0) and a mutant line, gigantea-2 (gi-2), which develops very large rosettes. Data obtained from growth analysis and gas exchange measurements was used to train a genetic programming algorithm to parameterize and test the above model. The relationship between leaf area and plant biomass was found to be non-linear and variable depending on carbon partitioning. The model output was sensitive to the rate of photosynthesis but more sensitive to the amount of carbon partitioned to growing thicker leaves. The large rosette size of gi-2 relative to that of Col-0 resulted from relatively small differences in partitioning to new leaf area vs. leaf thickness.
