To elucidate the pathological roles of staphylococcal enterotoxin C (SEC) in bovine staphylococcal mastitis, a histopathological analysis of SEC-inoculated mammary glands was performed. SEC-inoculated mammary glands exhibited interstitial inflammation, and the leukocytes that migrated into the gland were predominantly polymorphonuclear neutrophils (PMN). In the gland cistern tissues dissected from SEC-inoculated mammary glands, epithelial cellular degeneration was observed. We also investigated the physiological effects of SEC on PMN in vitro. PMN migration was induced by culture supernatant of SEC-stimulated peripheral blood mononuclear cells (S-PBMC sup) but not by that of nonstimulated PBMC (N-PBMC sup). The concentration of interleukin-8 was significantly (P < 0.05) higher in S-PBMC sup than N-PBMC sup, and a significantly (P < 0.05) higher mRNA expression of growth-regulated oncogenes was detected in SEC-stimulated PBMC than in nonstimulated PBMC. Milk PMN collected from SEC-inoculated mammary glands produced more than 2 times the amount of superoxide at 1 day postinoculation (dpi) than at 0 dpi in the presence of phorbol 12-myristate 13-acetate (PMA). PMN cultured with S-PBMC sup for 24 h also produced significantly (P < 0.05) larger amounts of superoxide than those cultured with N-PBMC sup in the presence of PMA. Moreover, S-PBMC sup induced the long-time survival of PMN. These results indicate that SEC induces the activation of PMN via the stimulation of mononuclear cells.

Dromedary camel (Camelus dromedarius) can survive and produce considerable amount of milk during recurrent and prolonged hot and dry environment. Camel milk considered one of the most valuable food sources due to its nutritional value. Intramammary infection and other low hygienic measurers are the main factors that undermined the camel mammary gland welfare. Available studies elaborated in detail the role of innate factors like Peptidoglycan recognition protein and Lactoferrin in camel milk as potent antimicrobial factors. Despite the wide studies on camel antibodies, their role in the mammary gland immunity was scarcely addressed. The major lack of information about the immune system of camel mammary gland is of great setback in improving the camel dairy industry. This review has evaluated the available data on different aspects of the mammary immune system. The available data unfortunately are of general profile, which created wide gaps in understanding the camel mammary gland immune system in health and disease. Several measures were proposed as necessary approaches to drive considerable interests of wide and in depth research on the camel mammary gland immune system.In conclusion, the available knowledge on the immune system of camel mammary gland suffers from major lack in the role of the immunoglobulins in health and mastitis. Although, acceptable progress was made on defining the cellular populations of the mammary gland, their main activity during the infection needs to be revealed.

-The mammary gland performs a variety of immunological functions, including protecting itself from mastitis and protecting neonates from infectious agents. Several molecules that mediate lymphocyte trafficking in the immune system are also expressed in the mammary gland. This review is focused on the immunological function of these molecules, especially glycosylation-dependent cell adhesion molecule-1 (GlyCAM-1) and mucosal addressin cell adhesion molecule-1 (MAdCAM-1) in the mammary gland. GlyCAM-1 is expressed in the lactating mouse mammary gland. Endothelial cells produce this protein and secrete it into milk. The glycosylated modification of mammary gland GlyCAM-1 is different from that of the lymph nodes, and lacks the binding ability for L-selectin on lymphocytes. GlyCAM-1 in the mammary gland is not involved in lymphocyte migration, and probably has another function besides that of the lymph nodes. MAdCAM-1 is expressed on endothelial cells of small venules around mouse mammary lobules during lactation. This molecule has the ability to interact with α 4 ß 7 integrin on lymphocytes and mediates lymphocyte recruitment to the mammary gland. The density of ß 7 + /CD3 + T-cells is correlated with the density of the MAdCAM-1-stained area, suggesting that MAdCAM-1 may mediate the migration of these cells. In contrast, there is no relationship between MAdCAM-1 expression and the number of ß 7 + /c-IgA + B-cells, implying that some other factor is involved in lymphocyte migration to the mammary gland. Chemokines, such as IL-8, GRO-α, MCP-1, RANTES and MEC, have been detected in human and mouse mammary glands.
