The RNA3 of alfalfa mosaic virus (A1MV) encodes the P3 protein and the viral coat protein (CP). RNA3 molecules transcribed in vitro replicated in protoplasts and plants when inoculated in mixtures with AIMV RNA 1, RNA2 and CP. Transcripts with a deletion or inversion in the P3 gene replicated well in protoplasts but not in transgenic plants transformed with the P3 gene. Transgenic plants expressing the CP gene became infected after inoculation with a mixture of RNA 1, RNA2 and wild-type RNA3 transcripts without addition of CP to the inoculum. Transcripts with a deletion in the CP gene replicated at a reduced level in protoplasts but not in CP-transformed plants. This suggests that P3 and CP are both required for cell-tocell spread of AIMV and that mutations in the inoculum RNA could not be complemented in trans by the wild-type chimeric nuclear genes.

RNA 3 of alfalfa mosaic virus (AIMV) encodes the movement protein P3 and the viral coat protein which is translated from the subgenomic RNA 4. The 5'-leader sequences of RNA 3 of AIMV strains S, A, and Y differ in length from 314 to 392 nucleotides and contain a variable number of internal control regions of type 2 (ICR2 motifs) each located in a 27 nt repeat. Infectious cDNA clones were used to exchange the leader sequences of the three strains. This revealed that the leader sequence controls the specific ratio in which RNAs 3 and 4 are synthesized for each strain. In addition, it specifies strain specific differences in the kinetics of P3 accumulation in plants. Subsequent deletion analysis revealed that a 5'-sequence of 1 12 nt containing one ICR2 motif was sufficient for a 10 to 20% level of RNA 3 accumulation in protoplasts and a delayed accumulation in plants. Expression of alfalfa mosaic virus coat protein gene confers cross-protection in transgenic tobacco and tomato plants

A chimeric gene encoding the alfalfa mosaic virus (AlMV) coat protein was constructed and introduced into tobacco and tomato plants using Ti plasmid-derived plant transformation vectors. The progeny of the self-fertilized transgenic plants were significantly delayed in symptom development and in some cases completely escaped infection after inoculated with AIMV. The inoculated leaves of the transgenic plants had significantly reduced numbers of lesions and accumulated substantially lower amounts of coat protein due to virus replication than the control plants. These results show that high level expression of the chineric viral coat protein gene confers protection against AIMV, which differs from other plant viruses in morphology, genome structure, gene expression strategy and early steps in viral replication. Based on our results with AIMV and those reported earlier for tobacco mosaic virus, it appears that genetically engineered crossprotection may be a general method for preventing viral disease in plants.
