Vardenafil reverses erectile dysfunction induced by paroxetine in rats

Abstract: Selective serotonin reuptake inhibitors (SSRIs) are associated with a high incidence of impotence. Paroxetine is an extensively used SSRI that has been shown to impair erectile function in patients, to induce erectile dysfunction and to inhibit nitric oxide synthase (NOS) activity and NO production in animal models. NO is a key mediator of penile erection. Vardenafil is a type 5 phosphodiesterase inhibitor that potentiates NO-mediated responses in isolated trabecular smooth muscle and penile erection in men in… Show more

“…Additionally, SSRI-induced erectile dysfunction was measured using intracavernous pressure assessments. Phosphodiesterase 5 inhibitor (PDE-5) inhibitors such as vardenafil and DA-8159 have been shown to reverse SSRI-induced decreases in intracavernous pressure (Ahn et al 2005;Angulo et al 2003). These preclinical effects are consistent with the observed clinical improvements in sexual function when sildenafil is combined with SSRIs (Nurnberg 2001;.…”

“…This hypothesis is supported by the evidence that acute or chronic paroxetine, but not citalopram, caused ED in rats by inhibiting NO production [139]. Indeed, the inhibitory effects induced by acute paroxetine on erectile function in the rat can be prevented by inhibition of PDE5 with vardenafil [140] or by coadministration of the NOS substrate, L -arginine [141]. On the other hand, venlafaxine, a mixed inhibitor of serotonin and norepinephrine (NE) reuptake, produced ED in rats by increasing NE levels, since its inhibitory effects on erectile responses were prevented by phentolamine [141].…”

“…The significant (p < 0.05) increase in ML and IL, an indicator of sexual arousability and potency, in sexually impaired rats could have resulted from reduced erectile response through the inhibition of the synthesis of nitric oxide in the penis leading to reduced carvenous stimulation in paroxetine-treated rats [27][28][29]. The capability of the extract to reverse the increase in ML and IL of sexually impaired rats indicates stimulation of sexual arousability, performance, motivation, and vigor in the rats [30].…”
