Vanillic Acid Stimulates Anagen Signaling via the PI3K/Akt/ β-Catenin Pathway in Dermal Papilla Cells

Abstract: Hair loss is a distressing disorder associated with various factors, including hormone imbalance, autoimmune diseases, stress, and chemotherapy. Several known types of hair loss are androgenetic alopecia (AGA), telogen effluvium, alopecia areata, and chemotherapy-induced alopecia (Cotsarelis and Millar, 2001; Hunt and McHale, 2005; Ito, 2010). Minoxidil (Rogain ®) and finasteride (Propecia ®) have been approved by the Food and Drug Administration (FDA) for treatment of alopecia. Minoxidil was developed as a va… Show more

“…Low-level laser therapy (LLLT) also seems to improve hair regrowth in the conditions of AGA, AA, and chemotherapy-induced alopecia [ 133 ]. There are studies demonstrating the positive effects of natural bioactive compounds on AGA [ 24 , 30 , 52 , 55 , 56 , 58 , 134 , 135 , 136 , 137 , 138 ]. A recently published study demonstrates that a 5-mer peptide (GLYYF; P5) has the potential to promote hair growth when topically applied [ 139 ].…”

“…These results demonstrated that BDB can effectively increase DPC proliferation. Cell proliferation and death are associated with changes in cell cycle-related proteins [ 27 , 34 ]. To investigate the mechanism by which BDB promotes DPC proliferation, we examined the levels of cell cycle-related proteins in DPCs.…”

“…Hair growth is positively or negatively regulated by various signaling factors. The mechanisms by which BDB promotes hair growth and proliferation of DPCs in this study are as follows: First, the Wnt/β-catenin pathway plays an important role in hair growth as well as in the regulation of proliferation of DPCs [ 22 , 27 , 36 ]. It has been reported that the Wnt/β-catenin pathways are involved in hair regeneration after wounding, prolongation of the anagen phase, and inhibition of apoptosis in DPCs [ 22 , 35 ].…”

“…All hair follicles periodically undergo a hair cycle that consists of anagen (growth), catagen (regression) and telogen (rest) ( Ji et al, 2021 ). Many recent studies have reported that hair cycle is regulated by the interaction between mesenchymal cells and epithelial cells in hair follicles, and the proliferation of dermal papilla (DP) cells by the activation of PI3K/Akt/Wnt/β-catenin pathway may be the key factor ( Botchkarev and Kishimoto, 2003 ; Kang et al, 2020 ).…”
