Value of Podoplanin as an Immunohistochemical Marker in Tumor Diagnosis

Nelson G. Ordóñez

Abstract: Podoplanin is a type I integral membrane glycoprotein that, because it is expressed in lymphatic endothelium, but not in vascular blood vessel endothelial cells, is commonly used in the identification of lymphatic endothelial differentiation in vascular endothelial neoplasms and lymphatic invasion by tumor. Because podoplanin is also expressed in mesothelial cells and fetal gonocytes, it has proved to be a useful marker for assisting in the differential diagnosis of mesotheliomas and germ cell tumors, particul… Show more

“…CLEC-2 binds to mucin glycoprotein podoplanin [378-380]. Podoplanin is expressed on cells of the lymphatic endothelia, type I lung epithelia, choroid plexus epithelia, kidney podocytes, lymph node stroma, along with cancer-associated fibroblasts (CAF) and tumor cells where it is found at the leading edge and is thought to potentiate migration and invasion [381, 378, 382, 383, 379, 380, 384]. Podoplanin is a heavily O -glycosylated type-1 transmembrane sialomucin glycoprotein [378, 357, 385, 386].…”

“…Another subset of heterotypic receptor interactions can potentially contribute to TCIPA when mucin-producing tumor cells are involved. In some instances, platelet CLEC-2 can bind to tumor cell podoplanin during heterotypic aggregate formation [378, 382, 383, 379, 380]. In other cases, P-selectins are also involved when mucin-producing tumor cells enter the blood stream and initiate TCIPA [475-479].…”

“…Podoplanin, a type I integral membrane glycoprotein, is a highly sensitive and specific marker for lymphatic endothelial cells, because it is expressed in lymphatic endothelium, but not in vascular blood vessel endothelial cells. Podoplanin is commonly used to identify lymphatic endothelial differentiation in vascular endothelial tumors and to detect lymphatic invasion by tumor . In further studies on various other tumor entities (eg, head and neck squamous cell carcinoma), podoplanin served as a marker for LVI with significant prognostication of outcome …”

[30][31][32][33][34][35][36] The purpose of this study was to report tumor characteristics and clinical course with regard to possible predictive parameters of head and neck mucosal melanoma seen at a single skin and head and neck cancer center and to assess the prognostic value of S100/podoplanin and S100/ CD31 double immunostaining as markers for LVI and blood vessel infiltration (BVI) by head and neck mucosal melanoma tumor cells.…”
