Valproic Acid Inhibits Leptin Secretion and Reduces Leptin Messenger Ribonucleic Acid Levels in Adipocytes

Abstract: Treatment of epilepsy or bipolar disorder with valproic acid (VPA) induces weight gain and increased serum levels for the satiety hormone, leptin, through an unidentified mechanism. In this study we tested the effects of VPA, a short-chain branched fatty acid (C8:0), on leptin biology and fatty acid metabolism in 3T3-L1 adipocytes. VPA significantly reduced leptin secretion in a dose-dependent manner. Because fatty acid accumulation has been hypothesized to block leptin secretion, we tested the effect of VPA o… Show more

“…Mouse Adipocyte Cell Culture-3T3-L1 preadipocytes were obtained from the American Tissue Culture Collection (Manassas, VA) and grown according to standard protocols (36). Confluent preadipocytes were differentiated in adipocyte media (DMEM, 10% FBS, 850 nM insulin, and 0.1% penicillin/ streptomycin) supplemented with 250 nM dexamethasone and 100 M isobutylmethylxanthine (IBMX) for 3 days.…”

“…Between 2 and 30 min thereafter, media were removed, and 100 l of 1ϫ Laemmli buffer was added to stop the reaction and lyse the adipocytes. Oil Red O staining of adipocytes and quantification of extracted dye were carried out as described previously (36).…”

“…Recently, VPA was proposed to induce a state of leptin resistance, much like obesity, leading to increases in appetite and body weight [1, 5]. At the molecular level, VPA was shown to inhibit histone deacetylase (HDAC) activity [6, 7], but its metabolic actions could be due to its inhibitory effects on the adipogenic transcription factor CCAAT enhancer binding protein α (CEBPα) [8, 9]. Elucidating the mechanisms of VPA-induced weight gain could help develop more effective antiepileptic therapies, but might also provide insight into the development of metabolic imbalances and the etiology of obesity.…”

“…19 Conversely, adipogenic agents like the thiazolinediones and valproic acid reduce leptin secretion. [20][21][22] Furthermore, adipocyte-selective reduction of leptin receptors leads to adiposity in mice, despite a normal level of hypothalamic leptin receptors and normal food intake. 16 These findings suggest adipogenic role of hypoleptinemia.…”
