Abstract: A novel, simple and economic reverse phase High Performance Liquid Chromatography (RP-HPLC) method has been developed for the quantification of Imatinib in bulk and capsule dosage form with greater precision and accuracy. Separation was achieved on Analytical technologies, C-18, (250mm*4.6mm) column in isocratic mode with mobile phase consisting of acetonitrile: potassium dihydrogen phosphate buffer (pH 2.5) (30:70v/v) with a flow rate of 0.8 mL/min. Hyperthyroidism is a condition characterized by increased in synthesis and secretion of thyroid hormones from thyroid gland. Methimazole is an anti-hyperthyroidism drug. It is of great importance and significance for life science. Up to now, various different methods have been applied for the determination of methimazole, such as spectrophotometry1-3, LC4, HPLC6, RP-HPLC7, HPLC-UV8, electrochemical analysis9, kinetic method10. In this paper, a new UV-Spectrophotometric method for the determination of methimazole has been established. The chemical name of Methimazole is (1-methylimidazole-2-thiol). The chemical formula of Methimazole is C4H6N2S and the molecular weight is 114.17 g/mol. The structure of Methimazole features a 5 membered imidazole ring with a methyl group at position 1st and thione group at position 2nd. Methimazole is white colour powder and has melting point 1440 C.

Simple sensitive and accurate extractive colorimetric method was developed for the estimation of Imatinib mesylate in Pharmaceutical dosage forms. The method was based on the formation of colored ion pair complexes by the drugs with thiocynate ions. These ion pair complexes were quantitatively extracted under the experimental condition in chloroform. The absorbance values were measured at 618 respectively. The proposed method was validated statistically. A recovery of method was carried out by standard addition methods. The Beer’s law ranges were found to be 1-12μg/ml, respectively. The low values of standard deviation and percentage RSD indicate high precision of method. Hence the method is useful for routine estimation of Imatinib mesylate in tablets respectively.

A Simple and precise UV-spectroscopic method was developed for the determination of Imatinib Mesylate in its bulk and formulation. The developed method has been validated for various parameters like specificity, accuracy, linearity, robustness according to USP general chapter<1225> and ICH Q2R1 guidelines. Pure solution of Imatinib Mesylate was scanned in the whole range of UV region where it has shown the maximum absorbance at 258nm. The RSD values for method precision and intermediate precision were found to be well within the acceptance criteria and the series of dilutions were found to be linear (2-12ug/ml) where r2= 0.999 was the regression value. Limit of detection (0.2925μg/ml) and Limit of quantification (0.8977μg/ml) of IMT were established. Further, the drug has been subjected to various stress conditions, and percent degraded was reported. Drug solutions have shown stability on the benchtop for up to 8 hours and 72 hours when the solutions were refrigerated.
