Validation of an enzyme-linked immunosorbent assay for the quantification of human IgG directed against the repeat region of the circumsporozoite protein of the parasite Plasmodium falciparum

Abstract: BackgroundSeveral pre-erythrocytic malaria vaccines based on the circumsporozoite protein (CSP) antigen of Plasmodium falciparum are in clinical development. Vaccine immunogenicity is commonly evaluated by the determination of anti-CSP antibody levels using IgG-based assays, but no standard assay is available to allow comparison of the different vaccines.MethodsThe validation of an anti-CSP repeat region enzyme-linked immunosorbent assay (ELISA) is described. This assay is based on the binding of serum antibod… Show more

“…Anti-circumsporozoite antibodies were measured by means of enzyme-linked immunosorbent assay 15 in the first 200 infants enrolled at each study center at screening and 1 month after dose-3. An antibody titer of 0.5 EU per millimeter or greater was considered to be positive.…”

“…Other than the reagents, operation or environmental factors, we also need to verify whether the ingredients of sample would affect the detection result [5] , which is required by Chinese Pharmacopoeia. If so, we would be able to observe such nonspecific disturbance effect in other ELISA tests.…”
