Background Nicotine administration can generate severe oxidative stress and lipid peroxidation. Royal jelly, with its antioxidant properties, acts as a scavenger of reactive oxygen species. This study describes the apitherapy effects of royal jelly on testicular damage following nicotine administration. Methods Forty‐eight male BALB/c mice were divided into 8 groups ( n = 6): saline, 3 different doses of royal jelly (100, 150, and 200 mg/kg body weight (BW) per day), nicotine (1.5 mg/kg), and 3 different groups of Nic + Roy (1.5 mg/kg of Nic + 100, 150, and 200 mg/kg BW per day of royal jelly). Nicotine was administrated intraperitoneally, and royal jelly was prescribed orally for 10 consecutive days. Serum levels of hormones (testosterone, luteinizing hormone, and follicle‐stimulating hormone), total antioxidant capacity, nitric oxide (NO) status, malondialdehyde levels, sperm DNA fragmentation, sperm parameters, histopathological changes (H&E staining), immunohistochemistry against apoptotic proteins, and gene expression of Bcl ‐ 2 , p53 , Caspase ‐ 3 , and Nrf2 (real‐time PCR) were assessed to evaluate the molecular and histological changes. Results Hormone levels, sperm parameters, and status of antioxidants were decreased significantly ( p < .05) following nicotine administration. Moreover, royal jelly treatment normalized hormonal and antioxidant characteristics, decreased apoptotic gene expression, increased Nfr2 gene expression, and restored histopathological alteration to the physiological status significantly ( p < .05). Conclusion Royal jelly upregulates the antioxidant status, inhibits the mitochondrial‐dependent apoptosis pathway, and increases the rate of proliferation. This therapeutic agent effectively protected the testis against nicotine‐associated damages by antioxidant and anti‐apoptotic effects.

Anti-Mullerian hormone (AMH) appears to be the best biochemical-endocrine marker in assessing the age-related decline of the ovarian follicular reserve. There is inadequate information on age-related changes in peripheral AMH concentrations in goats (Capra hircus). The present study thus aimed (i) to investigate the responsiveness of the ovary to gonadotrophin releasing hormone (GnRH) challenge to release plasma AMH into circulation in view of addressing the question whether ovarian secretion of AMH would be affected by gonadotrophic status in goats and (ii) to determine plasma AMH concentrations at different ages in goats (n= 82) using enzyme immune-assay technique. Data on hormonal concentrations in GnRH-treated animals as well as in different ages of animals were analyzed by nonparametric one-way repeated measure ANOVA and one-way ANOVA, respectively. There was no change (P>0.05) of plasma AMH concentrations after GnRH challenge in experimental goats. Plasma AMH concentrations changed significantly (P<0.05) between the mean rank of the related groups over the time (age) in goats. Plasma AMH concentration at 3 months of age was 0.36 ± 0.04 ng/ ml and thereafter it started increasing and reached  2 ng/ ml between 3 and 4 years of age and then showed a progressive decline from 5 years of age and reached at 0.30 ± 0.07 ng/ ml at 10 years of age. A cubic model was found to be the best fitted model that delineated the change of plasma AMH level with age. Hence, the present study indicated that peripheral AMH concentration could be a useful management tool for the selection of goats in breeding programmes.

Life science areas require specific sample pretreatment to increase the concentration of the analytes and/or to convert the analytes into an appropriate form for the detection and separation systems. Various workstations are commercially available, allowing for automated biological sample pretreatment. Nevertheless, due to the required temperature, pressure, and volume conditions in typical element and structure-specific measurements, automated platforms are not suitable for analytical processes. Thus, the purpose of the presented investigation was the design, realization, and evaluation of an automated system ensuring high-precision sample preparation for a variety of analytical measurements. The developed system has to enable system adaption and high performance flexibility. Furthermore, the system has to be capable of dealing with the wide range of required vessels simultaneously, allowing for less cost and time-consuming process steps. However, the system's functionality has been confirmed in various validation sequences. Using element-specific measurements, the automated system was up to 25% more precise compared to the manual procedure and as precise as the manual procedure using structure-specific measurements.
