Vaccinia Virus Blocks Gamma Interferon Signal Transduction: Viral VH1 Phosphatase Reverses Stat1 Activation

Abstract: We have analyzed the effects of vaccinia virus (VV) on gamma interferon (IFN-␥) signal transduction. Infection of cells with VV 1 to 2 h prior to treatment with IFN-␥ inhibits phosphorylation and nuclear translocation of Stat1 and consequently blocks accumulation of mRNAs normally induced by IFN-␥. While phosphorylation of other proteins in the IFN-␥ pathway was not affected, activation of Stat1 by other ligandreceptor systems was also blocked by VV. This block of Stat1 activation was dose dependent, and altho… Show more

“…Both VH1 and VHR differ from other DSPs in that they are much smaller (19 and 21 kDa, respectively). VH1 has been reported to dephosphorylate both MAP kinases and STAT1 (Najarro et al, 2001), while VHR appears to be specific for ERK and JNK (Denu and Dixon, 1995;Alonso et al, 2001). LMW-DSP2 was found to dephosphorylate and deactivate p38 MAPK and JNK, but not ERK .…”

“…For example, myxoma virus has been shown to secrete a homolog of the IFN-c receptor, which inhibits the biological activity of extracellular . Similarly, vaccinia virus encodes a soluble IFN-a/b receptor [18], and the viral phosphatase VH1 blocks IFN-c signaling [19]. The E1A protein of adenovirus blocks IFN responses by interfering with transcription and also directly suppresses Stat1 function [20].…”

“…The first DSP to be cloned, the VH1 protein from the vaccinia virus (23), is a member of this group, as are the human VHR (24) and a number of genes given the genomic designations DUSP14, DUSP19, DUSP20, DUSP21, and DUSP22. Although VH1 has been reported to dephosphorylate both MAP kinases and Stat1 (25), and VHR can dephosphorylate Erk and Jnk in 293T cells (26) and T cells (27,28), it appears that at least some of these small atypical DSPs also have functions related to MAP kinases.…”
