Recently, the use of phototrophs for wastewater treatment has been revisited because of new approaches to separate them from effluent streams. One manifestation uses oxygenic photogranules (OPGs) which are dense, easily-settleable granular biofilms of cyanobacteria, which surrounding populations of heterotrophs, autotrophs, and microalgae. OPGs can remove COD and nitrogenous compounds without external aeration. To better grow and maintain biomass in the proposed wastewater process, this study seeks to understand the factors that contribute to successful granulation. Availability of initial inorganic nitrogen, particularly ammonium, was associated with successful cultivation of OPGs. In the first days of granulation, a decrease in ammonium coupled with an increase in a cyanobacterial-specific 16S rRNA gene, may suggest that ammonium was assimilated in cyanobacteria offering a competitive environment for growth. Though both successful and unsuccessful OPG formation demonstrated a shift from non-phototrophic bacterial dominated communities on day 0 to cyanobacterial dominated communities on day 42, the successful community had a greater relative abundance (46%) of OTUs associated with genera Oscillatoria and Geitlernema than the unsuccessful community (27%), supporting that filamentous cyanobacteria are essential for successful OPG formation. A greater concentration of chlorophyll b in the unsuccessful OPG formation suggested a greater abundance of algal species. This study offers indicators of granulation success, notably availability of inorganic nitrogen and chlorophyll a and b concentrations for monitoring the health and growth of biomass for a potential OPG process.Electronic supplementary materialThe online version of this article (doi:10.1186/s13568-017-0444-8) contains supplementary material, which is available to authorized users.

It is well acknowledged that the activities of activated sludge (AS) are influenced by seasonal temperature variation. However, the underlying mechanisms remain largely unknown. Here, the activities of activated sludge under three simulated temperature variation trends were compared in lab-scale. The TN, HN3-H, and COD removal activities of activated sludge were improved as temperature elevated from 20 °C to 35 °C. While, the TN, HN3-H, COD and total phosphorus removal activities of activated sludge were inhibited as temperature declined from 20 °C to 5 °C. Both the extracellular polymer substances (EPS) composition (e.g., total amount, PS, PN and DNA) and sludge index of activated sludge were altered by simulated seasonal temperature variation. The variation of microbial community structures and the functional potentials of activated sludge were further explored by metagenomics. Proteobacteria, Actinobacteria, Acidobacteria and Bacteroidetes were the dominant phyla for each activated sludge sample under different temperatures. However, the predominant genera of activated sludge were significantly modulated by simulated temperature variation. The functional genes encoding enzymes for nitrogen metabolism in microorganisms were analyzed. The enzyme genes related to ammonification had the highest abundance despite the changing temperature, especially for gene encoding glutamine synthetase. With the temperature raising from 20 °C to 35 °C. The abundance of amoCAB genes encoding ammonia monooxygenase (EC:1.14.99.39) increased by 305.8%. Meanwhile, all the enzyme genes associate with denitrification were reduced. As the temperature declined from 20 °C to 5 °C, the abundance of enzyme genes related to nitrogen metabolism were raised except for carbamate kinase (EC:2.7.2.2), glutamate dehydrogenase (EC:1.4.1.3), glutamine synthetase (EC:6.3.1.2). Metagenomic data indicate that succession of the dominant genera in microbial community structure is, to some extent, beneficial to maintain the functional stability of activated sludge under the temperature variation within a certain temperature range. This study provides novel insights into the effects of seasonal temperature variation on the activities of activated sludge.

The application and development of environmental laws governing watershed management requires identification of biological indicators, parameters and measures to establish its ecological status, for which the present study aims to characterize the ecological indicator value of stream benthic algae in an urban area of Mexico City. Six sites of the Magdalena river were studied in the most contrasting seasons: drywarm, rainy and dry-cold. At each site the physicochemical parameters of water were recorded and 10 samples were collected: Five with visible algal growths and five with diatoms. The environmental quality of each site was determined by the trophic status and its confirmation with the richness and benthic algal diversity, through a canonical correspondence analysis and the calculation of the indicator value of species (IndVal). According to the trophic status, three groups of sites were recognized. The first group (G1) was assessed as reference of the oligotrophic conditions and presented 11 species with the highest IndVal, protruding: Encyonema silesiacum, Gomphonema parvulum, Navicula cryptocephala, Planothidium lanceolatum, Vaucheria bursata, Placoma regulare and Nostoc parmelioides. A second group (G2) was associated with 15 species considered detectors, which were tolerant to increased nutrients in at least one season, such as Diatoma mesodon and Prasiola mexicana. The third group (G3) included sites with species that had a wide range of tolerance to pollution: Fistulifera saprophila, Nitzschia minuta, Nitzschia palea, Nitzschia recta and Phormidium autumnale. The species IndVal of Magdalena River has established reference sites of good environmental quality and those that are altered by human activities, therefore they are recommended as the first step in a methodological proposal for a long-term biomonitoring through a biological index.
