UV Fluorescence-Based Determination of Urinary Advanced Glycation End Products in Patients with Chronic Kidney Disease

Abstract: Advanced glycation end products (AGEs) are a class of proteins or lipids that are non-enzymatically glycated and oxidized after contact with aldose sugars. The accumulation of AGEs results in carbonyl stress, which is characteristic for diabetes mellitus, uremia, atherosclerosis and vascular dysfunction. In recent decades, several innovative methods have been developed to measure the concentration of AGEs in blood or urine. In the present study, we evaluated the use of UV fluorescence as an alternative tool to… Show more

“…As described previously [ 24 , 45 ], AGEs were quantified based on Maillard-type autofluorescence (AF) measurements (excitation 365 nm, emission 390–700 nm) using a Flame miniature spectrometer (FLAME-S-VIS-NIR-ES, 350–1000 nm, Ocean Optics, Dunedin, Florida, USA) equipped with a high-power LED light source (365 nm, Ocean Optics) and a reflection probe (QR400-7-VIS-BX, Ocean Optics). Measurements were averaged over 128 scans.…”

“…Since fluorescence spectroscopy is a valuable and commonly employed method for the detection and measurement of autofluorescent AGEs [ 17 , 18 ], Maillard-type AF measurements (excitation 370 nm, emission 390–700 nm) were performed using a Flame miniature spectrometer (FLAME-S-VIS-NIR-ES, 350–1000 nm, Ocean Optics, Dunedin, FL, USA) equipped with a high-power LED light source (365 nm, Ocean Optics) and reflection probe (QR400-7-VIS-BX, Ocean Optics). “…The interpretation of the second peak is not so obvious. Our proposition about the AGE products is based on preliminary experiments with the fluorescent spectra of the various chromatographic fractions of hemodialysate and AGE products observed during HD [66][67][68][69]; however, additional comprehensive research is needed to fully support this suggestion. The clinical significance of the second and the third peaks in the chromatograms is yet to be investigated; the second peak is of special interest because AGE products are responsible for cardiovascular complications and other pathological conditions in PD patients [70][71][72].…”
