UV‐B photoreceptor UVR8 interacts with MYB73/MYB77 to regulate auxin responses and lateral root development

Abstract: The UV-B photoreceptor UVR8 mediates multiple UV-B responses in plants, but the function of UVR8 in regulating root development has not previously been investigated. Here, we show that UV-B light inhibits Arabidopsis lateral root growth in a UVR8-dependent manner. Monomeric UVR8 inhibits auxin responses in a tissueautonomous manner and thereby regulates lateral root growth. Genome-wide gene expression analysis demonstrated that auxin and UV-B irradiation antagonistically regulate auxin-regulated gene expressio… Show more

“…The expression of UVR8 is also found in roots along with stems and leaves (Rizzini et al 2011). MYB transcription factors MYB73/77 regulate lateral root growth by interacting with UVR8 in roots in presence of UV-B by inhibiting auxin signaling and hence integrating UV-B and auxin signaling pathways (Yang et al 2019).…”

“…Current models of UVR8 signalling involve the monomer binding to COP1, which results in stabilisation of HY5 protein (Favory et al, 2009;Rizzini et al, 2011;Huang et al, 2013), which in turn can stimulate downstream responses including its own transcription (Binkert et al, 2014). Signalling also involves direct interaction of UVR8 monomers with specific transcription factors to initiate downstream responses (Liang et al, 2018;Yang et al, 2018Yang et al, , 2020. However, there is no evidence that monomer photoreception is more, or less, likely to initiate signalling than dimer photoreception.…”

“…The transcription factors BIM1 and BES1 mediate brassinosteroid (BR) signalling and stimulate extension growth; interaction with UVR8 in the nucleus reduces binding of these transcription factors to BR-responsive genes and hence promotes hypocotyl growth suppression by UV-B (Liang et al, 2018). MYB73 and MYB77 regulate genes involved in auxin-controlled responses, several of which are inhibited by UV-B (Vanhaelewyn et al, 2016;Jenkins, 2017); physical interaction of monomeric UVR8 with these transcription factors impairs their promoter binding activity and hence inhibits auxin-stimulated lateral root growth (Yang et al, 2020).…”

“…In addition to HY5, the transcription factors HY5 HOMOLOG (HYH), FAR‐RED ELONGATED HYPOCOTYLS 3 (FHY3), BES1‐INTERACTING MYC‐LIKE1 (BIM1), WRKY36, and MYB DOMAIN PROTEIN 73 and 77 (MYB73/MYB77) participate in the regulation of gene expression in the UVR8 pathway (Brown and Jenkins 2008; Huang et al 2012; Liang et al 2018; Yang et al 2018; Liang et al 2019; Yang et al 2019). Interestingly, recent work has unraveled several UV‐B‐responsive genes that are not regulated by UVR8, suggesting the existence of alternative UV‐B signaling pathway(s) (O'Hara et al 2019).…”
