Using prodigiosin against some gram-positive and gram-negative bacteria and Trypanosoma cruzi

Abstract: Background: This work aimed to explore the action of natural prodigiosin on both bacterial organisms and Trypanosoma cruzi cells. Methods: Natural prodigiosin pigment was extracted and purified from cultures of Serratia marcescens . Two media, peanut broth and peptone glycerol broth, both recommended in the literature for prodigiosin production, were compared. The prodigiosin obtained was employed to explore its anti… Show more

“…Arivizhivendhan et al reported the inhibitory activity of S. marcescens produced against P. aeruginosa [ 93 ]. Similar results of antibacterial activities by S. marcescens 2170 PG have also been reported by Herráez et al [ 94 ]. PG produced by S. marcescens UFPEDA 398 showed inhibitory activity against 20 strains of oxacillin-resistant S. aureus .…”

“…The antagonist effect of S. marcescens 2170 produced PG has been tested against parasite Trypanosoma cruzi cells. The results showed that the concentration of PG required to suppress T. cruzi growth was significantly lower than that required for benznidazole (0.25 mg/L and 4.9 mg/L, respectively) [ 94 ]. Isaka et al reported three compounds (metacycloprodigiosin, bafilomycin A 1 , and spectinabilin) extracted from Streptomyces spectabilis BCC 4785 possessed potent antimalarial activity against P. falciparum K1.…”

“…Our group recently studied prodigiosin of bacterial origin, reporting its remarkable activity against the epimastigotes of T. cruzi (CL strain, clone B5), with very low IC 50 values (up to 0.8 µM), compared to benznidazole (18 µM). Additionally, we explored the effects of this pigment on the parasite cell surface by atomic force microscopy, which revealed relevant morphological alterations in the size, shape, and roughness [ 26 ]. Based on these encouraging preliminary results, we decided to continue exploring the trypanocidal effect of prodigiosin and related molecules against the intracellular forms of T. cruzi and investigate the targets presumably involved.…”

“…without bicarbonate (Sigma-Aldrich, São Paulo, Brazil). Conidial suspensions were obtained from sporulated Aspergillus cultures and adjusted to contain 2-5×10 6 conidia/mL by counting in a hemocytometer. To obtain a final concentration of 2-5×10 5 conidia/mL, 1:10 dilutions were prepared in sterile distilled water.…”
