Usefulness of perflubutane microbubble-enhanced ultrasound in imaging and detection of prostate cancer: phase II multicenter clinical trial

Abstract: CEUS with Sonazoid(®) improved the detection rate of PCa by visualizing cancerous lesions. More detailed examination of CEUS images provided efficient characterization especially in the TZ area. TBx according to this procedure is expected to enable a lower number of biopsies and more accurate diagnosis of PCa.

“…However, it not clear whether the TIC values are useful for the precise assessment of cancer aggressiveness, which is commonly estimated by the Gleason score and largely influences PCa management2122. Moreover, the contrast-enhanced CETRUS findings for the PCa lesions showed various patterns according to tumour vascularity2324 and aggressiveness2526; this is indicative of the heterogeneity of this cancer. In our study, incorporating TICs enabled the differentiation of low-grade tumours (n = 23) from high-grade ones (n = 31) (Fig.…”

“…Microbubbles for diagnosis and treatment of breast, prostate, and liver cancer, among others, have been tested in clinical trials with promising results 24 . Sonazoid ® microbubbles were shown to improve the accuracy of diagnosis of prostate cancer, which could reduce the number of biopsies patients need to receive to achieve a similar diagnostic result 31 . In Japan, Sonazoid ® was also used in a phase III clinical trial to compare its diagnostic potential of focal breast lesions against unenhanced ultrasound and contrast enhanced MRI (Figure 2) 32 .…”

“…TBx had a positive overall detection rate of 30.9%, whereas the overall detection was only 6.9% with the 12-core TRUS-guided biopsies [2]. Nontargeted ultrasound microbubbles were studied by Uemura et al [3] who performed targeted prostate biopsies with microbubbles in 71 patients. Although there was a significant difference in cancer detection rate by TBx between two cohorts with PSA less than 10 ng/ml (22.9%) and PSA of at least 10 ng/ml (52.2%), the added value of CE-TRUS was relatively small [3].…”
