Usefulness of Nucleolar Organizer Region Staining in Gastric Myogenic Tumors: Correlation with Ploidy by DNA Flow Cytometry

Abstract: A silver-staining technique to identify nucleolar organizer region-associated proteins (Ag-NOR) was applied to 55 gastric myogenic tumors. The mean numbers of Ag-NOR in the nucleus were: leiomyoma (30 cases), 2.0 + 0.6 (mean + SD); low-grade leiomyosarcoma (11 cases), 3.0 + 0.7; and high-grade leiomyosarcoma (14 cases), 3.9 + 0.7. The Ag-NOR counts were compared with DNA ploidy as determined by flow cytometry in 46 tumors. The Ag-NOR counts were significantly different in the aneuploid leiomyosarcomas (4.1 + 0… Show more

“…Recently developed analyses such as DNA ploidy pat tern [19][20][21][22], BrdU [21,23], and PCNA [12][13][14][26][27][28][29][30] may be useful measures of malignant potential for a variety of tumors. Monoclonal antibody against PCNA reacts immunohistochcmically not only with fresh-frozen tissues but also with paraffin-enbedded materials [31], This enabled the retrospective evaluation of the proliferative activity of malignant tumor consid ering the intratumoral heterogeneity.…”

“…It has been suggested that the AgNOR number differs significantly between benign and malignant tumors, and this number has been DroDosed as a useful Darameter for determining have suggested that the AgNOR number is closely related to positive immunohistochemical staining for Kiand to the number of S-phase cells shown by flow cytometric DNA analysis. 18,19 Therefore, because this method might be useful for predicting tumor biological behavior, we assessed the clinicopathological and prognostic value of the AgNOR number for HCC in the present study.…”
