Use of VIRAZOLE to eradicate odontoglossum ringspot virus from in vitro cultures of Cymbidium Sw.

Abstract: Odontoglossum Ringspot Virus has been eradicated from Cymbidium Sw. through chemotherapy based on incorporation of ribavirin (VIRAZOLE ®) into the in vitro culture medium of protocorms. Applications of the virustatic agent for several consecutive subcultures freed protocorms of the virus. Acclimated plantlets regenerated from those protocorms are healthy as determinated by enzyme-linked immunosorbent assay (ELISA). No resurgence of virus occurred over a period of 5 years. Besides, trueness to type was total at… Show more

“…In fact, the principal target of antiviral activity of these molecules is the inosine monophosphate dehydrogenase, an enzyme that catalyzes the conversion of inosine 5'-monophosphate (IMP) in xantosine 5'-monophosphate and is able to alter the pathway for the production of guanosine mono-di-and triphosphate. “…Chemotherapy was applied to embryo axes dissected from garlic cloves under aseptic conditions in an sterile air flow cabinet, and inoculated in 120 ml baby food jars (5 explants per jar) containing 20 ml of propagation medium supplemented with a filter-sterilized solution of Ribavirin (Virazole or 1-b-D-ribofuranosil-1, 2, 4-triazole-3-carboxamide; 205 lM) (Cassells, 1987;Toussaint et al 1993). When the explants had developed a new leaf (10-30 mm in length), this new more external leaf was dissected together with a thin layer of tissue from the base of the explant.…”

“…In apple shoot cultures ribavirin [virazole], an analog of guanosine has been used to eliminate Apple Chlorotic Leaf Spot Virus [ACLSV] in combination with tissue culture [9,10], used it in Cymbidium cultures to eradicate Odontoglossum Ringspot Virus [ORSV]. Potato viruses have been eliminated in potato meristem culture by using 2,4-dioxohexahydrol,3,5-triazine [11], and potato stem cuttings [12].…”
