Use of serial analysis of gene expression to generate kidney expression libraries

Abstract: Chronic renal disease initiation and progression remain incompletely understood. Genome-wide expression monitoring should clarify mechanisms that cause progressive renal disease by determining how clusters of genes coordinately change their activity. Serial analysis of gene expression (SAGE) is a technique of expression profiling, which permits simultaneous, comparative, and quantitative analysis of gene-specific, 9- to 13-bp sequence tags. Using SAGE, we have constructed a tag expression library from ROP-+/+ … Show more

“…Because the kidney mass consists predominantly of proximal tubules, a significant fraction of tags are mapped to genes known to be expressed in proximal tubular epithelial cells. Particularly, the most abundant transcripts in normal kidney were kap and glutathione peroxidase 3 (GPx3), in agreement with previous data (El-Meanawy et al 2000).…”

“…For example, SAGE of mouse kidney revealed a preponderance of ion channels and mitochondrial enzymes, consistent with the role of the kidney in filtration and solute transport and the high-energy requirement for the same (El-Meanawy et al, 2000). Transcriptional profiling of the 100 most abundant SAGE tags derived from seedlings of the rice plant, Oryza sativa L., demonstrated a prevalence of prolamin, a storage protein expressed in seeds (Matsumura et al, 1999).…”
