Use of RAPD Markers to Document the Origin of the Intergeneric Hybrid xMargyracaena skottsbergii (Rosaceae) on the Juan Fernandez Islands

Abstract: Bands of random amplified polymorphic DNA (RAPDs) were used as markers to test the hypothesis that the species x Margyracaena skottsbergii, which is endemic to the island of Masatierra in the Juan Fernandez archipelago, represents an intergeneric hybrid between Acaena argentea and Margyricarpus digynus. Thirteen IO-mer primers produced 18 consistent species-specific bands for A. argentea and 27 for M. digynus. with all 45 bands present in the presumed hybrid x Margyracaena.A second species of Acaena on Masatie… Show more

“…RAPD markers having proven their efficacy in assessing hybrid origin in plants (Crawford & al., 1993;Marsolais & al., 1993), with a hybrid combining the bands present in its progenitors, it is significant that none of the subspecies appeared to share bands with the two others in a way suggestive of hybridization. Each subspecies shared approximately 50 % of its bands with each of the two others.…”

“…Ancient species of coast redwood have been called Sequoia aYnis, S. langsdorWi, S. abietina, or S. couttsiae; and giant sequoia as Sequoiadendron chaneyi, and S. reichenbachi. However, these ancient redwood species are slightly diVerent in their foliage from present day Sequoia and Sequoiadendron, respectively (Endo 1928;Axelrod 1959;Fields 1993;Sawyer et al 2000a).…”
