Use of Phosphonic Acid as a Generic Hapten in the Production of Broad Specificity Anti-Organophosphate Pesticide Antibody

Abstract: Phosphonic acid (trans-4-phosphono-2-butenic acid; TPB) was used as a generic hapten in order to generate broad specificity antibodies against a group of organophosphorus pesticides. The polyclonal antiserum showed, in an indirect enzyme-linked immunosorbent assay (ELISA) format, preferential binding toward pesticides containing unsaturated diethyl-phosphate functionalities rather than the equivalent thiophosphate or dimethyl structures. The level of detection in the ELISA using a heterologous system was inves… Show more

“…With the increasing of detection request for chemical residue, immunoassay of total and multi-residue was focused on gradually. So, with higher CRs, some immunoassay for a type of compounds is also expected and some class specific antibodies organophosphorus [20][21][22] or pyrethoid insecticides have been reported [23][24][25]. Therefore, it is of great use to study the effect of coating hapten structure on immunoassay specificity.…”

“…The phenyl has two functions. First, it can improve the capability of evoking specific immune response of immuno-hapten (Alcocer et al, 2000;Xu, Yamamoto, & Janda, 2004). Moreover, similar to the nitrofuran group, phenyl is an electron-withdrawing group.…”

“…These cells are harvested from the animal and fused to an immortal cell line to allow for continual production of the antibody of interest. Researchers have utilized this approach to produce thousands of different antibodies with specificity for a range of biological and chemical targets, including dyes, explosives, carcinogens, and pesticides for sensor applications [125][126][127]. Effective immobilization of the molecular recognition elements is critical for ensuring the efficacy of the sensing surface.…”
