Use of para-amino hippuric acid to measure blood flows through portal-drained-viscera, liver and hindquarters in sheep

Abstract: SUMMARYThe objectives of this study were (i) to evaluate a new surgical procedure to catheterize hepatic vessels as well as the posterior aorta and vena cava, (ii) to test a modification of a downstream dilution technique in order to measure blood flows through splanchnic tissues and hindquarters in rapid succession, and (iii) to improve the reliability of splanchnic blood flow measurements using paraamino hippuric acid (PAH). Ten adult ewes were used. The transhepatic surgical approach used proved successful;… Show more

“…In addition, an absence of significant changes in the contribution of the hepatic arterial to total venous blood flow with intake has been previously shown (Vernet et al, 2005). Lastly, results from Ortigues-Marty et al (1994) and Huntington (1982) have shown that the portal and hepatic blood flows can be measured with a relative standard deviation (r.s.d.) of 11% and 16% to 32%, respectively, using the marker dilution technique.…”

“…Blood catheters were inserted into the portal and hepatic veins as described previously (Ortigues-Marty et al, 1994;Savary-Auzeloux et al, 2010). The lambs were also fitted with a transit-time ultrasonic blood flow probe on the portal vein (Type A; Transonic Systems Inc., Ithaca, NY, USA), housed in individual stalls at 208C with continuous lighting and had ad libitum access to drinking water and a trace mineral salt block.…”

“…De outra forma, embora não haja um método padrão de medida de fluxo de sangue que possa ser utilizado para comparação, os resultados de FSV obtidos no presente estudo (Figura 1), assim como o seu grau de variação, são similares aos obtidos em outros estudos com ovinos (KATZ & BERGMAN, 1969;ORTIGUES et al, 1994). Não foi observada uma variação regular de FSV ao longo do tempo após a refeição, e os desvios-padrão em cada tempo foram relativamente altos.…”

“…The PAH concentration in whole blood was determined as described by Ortigues et al [16] after a deacetylation step through acid hydrolysis. The blood flow rate was calculated according to the indicator-dilution technique [9].…”
