Abstract: Rodrigo de Freitas lagoon is an eutrophic aquatic environment. The waters from the lagoon are released to the sea at Ipanema and Leblon beaches, through Jardim de Alah channel. In this work, the influence of these waters on the bacterial communities of these beaches was investigated. Eleven sampling stations were set between the lagoon and the beaches, and the samples were analyzed by molecular and microbiological parameters. PCR-DGGE of the DNA extracted from the samples was performed using rpoB primers. Prel… Show more

“…The view of eutrophication is biased from observations in few systems (eg., Taketani et al , 2003; Araujo and Hagler, 2011). This scientific discipline is still very young to marine systems and a broader view of costal eutrophication will consider how anthropogenic nutrient enrichment interact with other stressors such as species translocation, habitat loss, fishing, inputs of toxic contaminants, manipulation of freshwater flows, aquaculture, and climate change.…”

“…Extraction, PCR, and DGGE analysis of community DNA A 0.22-lm membrane containing material obtained after the filtration of 300 mL of water collected from each sampling location was cut in half, and one of the pieces was then cut into small pieces for DNA extraction with the FastDNA Ò SPIN Kit for Soil (BIO101, California, USA), as described by Taketani et al (2003).…”

“…Furtado et al 2001), generally using only cultivation-dependent or other conventional methods. As far as we know, there has been only one report of the use of molecular tools to study bacterial communities in urban lagoons (Taketani et al 2003). Several papers have demonstrated the influence of imported populations on an environment using molecular methods (Bergström and Jansson 2000;Crump et al 1999;Lindströn 1998;Schauer et al 2000), but all of them analysed temperate climate environments, which present different characteristics than tropical ones.…”
