Use of hybridoma-resistant variant cell lines to study H-2D b/FMR-specific cytotoxic T cells

Abstract: An H-2Db heterozygous tumor cell line and a variant subclone bearing a mutant gene product were used to analyze the H-2Db specificity of cytotoxic T lymphocytes (CTL) generated during a Moloney murine sarcoma virus (MSV) infection. When the mutant cells were used as targets for MSV-specific CTL, the amount of cell lysis, compared with that seen with the nonmutant parental cells, was drastically decreased. However, cells of the mutant clones remained susceptible to allogeneic CTL specific for the nonmutant H-2D… Show more

“…Somatic H-2 variants expressing structurally modified H-2 molecules, which differ from wild type in determinants defined by monoclonal antibodies (mAb) [4, H-2 determinants recognized by CTL. Chapdalaine et al [6] first reported about an H-2 mutant of a moloney murine sarcoma virus (MSV)-induced cell line of (BALB.B x BALB/c)F1 origin (H-2Kb'd,Db'd), which was isolated by cytotoxic selection with a Db-specific mAb. This H-2 variant was hardly lysed by H-2b-restricted, Friend, Moloney, Rauscher virus (FMR)specific CTL, whereas alloreactive H-2b-specific CTL did not discriminate it from wild type cells.…”
