Use of dual-flow bioreactor to develop a simplified model of nervous-cardiovascular systems crosstalk: A preliminary assessment

Abstract: Chronic conditions requiring long-term rehabilitation therapies, such as hypertension, stroke, or cancer, involve complex interactions between various systems/organs of the body and mutual influences, thus implicating a multiorgan approach. The dual-flow IVTech LiveBox2 bioreactor is a recently developed inter-connected dynamic cell culture model able to mimic organ crosstalk, since cells belonging to different organs can be connected and grown under flow conditions in a more physiological environment. This st… Show more

“…The late rise in VEGF-A seems, instead, to rely upon the PKCβII/HuR cascade acting at post-transcriptional level, which favors the expression of VEGF 164 , the member primary implicated in vascular hyperpermeability and proliferation. To this last regard, these results confirm our previous findings showing, both in vitro (Amadio et al, 2008;Amadio et al, 2012;Marchesi et al, 2020;Platania et al, 2020) and in vivo (Amadio et al, 2010), a post-transcriptional control of VEGF-A expression mediated by the RNA binding protein (RBP) ELAV/HuR.…”

“…In the drug delivery study, the choice of using a bioreactor was made because this system is modular, with chambers designed to be added in sequence or in parallel, thus simulating a multiple-organ system [21]. Furthermore, it is designed to be consistent with plates or transwells, thus allowing the use of standard protocols for in vitro procedures.…”

“…An advanced reactor system was used to perform in vitro NAC release from MLC (IVTech ® , Lucca, Italy) [18,21]. The system consists of a peristaltic pump, which creates the flow, connected with modular and transparent double-flow bioreactors named LiveBox2 (LB2).…”

“…Phonomimetic bioreactor was invented to examine the human vocal folds fibroblasts under inflammatory conditions and vocal fold diseases treatment with regenerative medicine and tissue engineering application [ 9 , 10 ]. A nanovibrational bioreactor for controlled osteogenesis of MSCs without chemical induction was also designed [ 15 , 69 , 73 ]. This bioreactor system [ 30 ] requires mechanical stimulation with a frequency of 1 kHz for osteogenesis.…”
