Use of a quadrupole linear ion trap mass spectrometer in metabolite identification and bioanalysis

Abstract: A new type of quadrupole linear ion trap mass spectrometer, Q TRAP TM LC/MS/MS system (Q TRAP TM ), was evaluated for its performance in two studies: firstly, the in vitro metabolism of gemfibrozil in human liver microsomes, and, secondly, the quantification of propranolol in rat plasma. With the built-in information-dependent-acquisition (IDA) software, the instrument utilizes full scan MS in the ion trap mode and/or constant neutral loss scans as survey scans to trigger product ion scan (MS 2 ) and MS 3 exper… Show more

“…With a conventional tandem MS, a product ion scan can only be performed in a second chromatographic separation and, in addition, the instrument is less sensitive than ion trap instruments [12,13]. Therefore, the combination of a tandem mass spectrometer and a sensitive ion trap in a single instrument is the instrument of choice for precursor ion scan (PreIS) or CNL combined with the acquisition of product ion mass spectra [14,15].…”

“…The AC RF voltage oscillates between the two hyperbolic metal end cap electrodes, if ion excitation is desired the driving AC voltage is applied to the ring electrode. This results in the formation of a plume of trapped ions which oscillate between forming a thin stretched plume and a short squashed plume between the ring and end cap electrodes (Xia et al, 2003). An example of possibly the most commonly employed ion trap that is applied to LC in metabolomic analyses is the Thermo-Finnigan LTQ TM (Linear Trap Quadrupole; Fig.…”

“…However, the purification and isolation of each metabolite are normally required for obtaining NMR spectra. 5), 8) The purification of a small amount of metabolites in vitro is a time-consuming process. Therefore, we decided to use liquid chromatography/nuclear magnetic resonance spectroscopy (LC/NMR) in elucidating the structure of hydroxylated-form metabolites of gemfibrozil.…”
