Use of a high‐throughput umu‐microplate test system for rapid detection of genotoxicity produced by mutagenic carcinogens and airborne particulate matter

Abstract: In the present study, we developed a rapid umu-microplate test system that uses the nitroreductase- and O-acetyltransferase-overproducing Salmonella typhimurium strain NM3009 and the O-acetyltransferase-overproducing S. typhimurium strain NM2009 to detect genotoxic activity in small volume samples. “…S. typhimurium strain NM3009 can overproduce O-AT and NR, which are suggested to catalyze the key steps in the mutagenic activation of nitroarenes (Oda et al, 1993). If NM3009 have more sensitive response than TA1535/pSK1002 in detecting the genotoxic activities of environmental sample in the absence of S9, it would be concluded that the genotoxic activity is at least partially due to the nitroarenes (Oda et al, 2004). NM2009 overproducing O-AT has a high response as NM3009 to a few nitroarenes, such as 1,8-dinitropyrene, 3,7-dinitrofluoranthene, which may be activated to mutagens mainly by O-AT rather than NR (Oda et al, 1993).…”
