Use of a Fibrin Preparation in the Engineering of a Vascular Graft Model

Abstract: A structured coculture of MFB and EC was obtained mimicking the formation of a vascular wall with preserved viability utilizing a fibrin preparation. Nutrition problems seem to limit the maximal extent of MFB in the matrix.

“…In fact, Aper et al found that ECs adhered to fibrin gels extremely well and formed near confluent monolayers that remained viable for up to 19 days. 3 In either case, more work needs to be done to more precisely determine the composition of the luminal surface and the origin of the enhanced adhesion, which may have implications for improving EC retention on traditional synthetic grafts as well as tissue-engineered grafts.…”

“…The unique potential of fibrin as a scaffold has been well documented. 2,9,13,19,23,33,37 The necessity to optimize the concentrations of fibrin scaffold components 7,14 and fibrinolytic inhibitors 15 has been highlighted previously. As a result, the seeding technique and the fibrin component concentrations used in this study were based on those previously optimized for tissue engineering vascular grafts.…”
