Use and abuse of the quasi-steady-state approximation

Abstract: The transient kinetic behaviour of an open single enzyme, single substrate reaction is examined. The reaction follows the Van Slyke-Cullen mechanism, a spacial case of the Michaelis-Menten reaction. The analysis is performed both with and without applying the quasi-steady-state approximation. The analysis of the full system shows conditions for biochemical pathway coupling, which yield sustained oscillatory behaviour in the enzyme reaction. The reduced model does not demonstrate this behaviour. The results hav… Show more

“…We want to illustrate by a simple example that this procedure is problematic and may have to be replaced by more careful reasoning. (Such observations have been made earlier; see Flach and Schnell [6].) The following reaction system describes a situation where the enzyme is formed by degradation of another substance (a proenzyme) and itself is subject to degradation.…”

“…Nevertheless, the fact that differences can be identified illustrates an advantage of automating the design procedure. It is also possible that automation could obfuscate poorly justified assumptions, such as in the application of the QSSAs [34]; so it would be important to incorporate rigorous checks for their validity.…”

“…The simplifications, for example, of quasi-steady state approximations have been used in a qualitative manner. We have stressed on physical significance at every step rather than following mathematical rigour and accuracy (Flach & Schnell, 2006;Pedersen, Bersani, & Bersani, 2008).…”
