Ursolic acid derivative UA232 evokes apoptosis of lung cancer cells induced by endoplasmic reticulum stress

Abstract: Context: Ursolic acid (UA), a natural product, shows a broad spectrum of anticancer effects. However, the poor bioavailability and efficacy of UA limit its clinical application. Objective: We developed novel analogues of UA with enhanced antitumor activities by the extensive chemical modification of UA. Materials and methods: We developed multiple compounds by structural modification of UA, and found that UA232 had stronger activity than UA. The effects of UA232 (0-50 lM) on inhibiting the proliferation of A54… Show more

“…Its anti-tumor effect is particularly prominent. The anti-tumor effect of UA has been reported in a variety of diseases, including breast cancer (31), leukemia (32), prostate cancer (33), melanoma (34), endometrial cancer (35), and lung cancer (36), among others. A large number of studies have shown that UA can inhibit tumor growth and metastasis by inhibiting tumor cell proliferation, invasion, and metastasis, and its mechanism is related to the expression of related genes on tumor cells, the oxidation of fatty acids increases, and the ability to synthesize fatty acids decreases.…”

“…UA232 treatment increased expression of CHOP, indicating a mechanism involving the ER stress pathway. Pretreatment with 4-PBA, an ER stress inhibitor, attenuated the UA232-induced apoptosis, a further indication of the role of ER stress [ 88 ].…”

“…Our research group carried out a series of modifications to UA and screened out the derivative, UA232, that shows strong anti-tumor activity 17 . It was found that its anti-tumor effect on lung cancer was significantly stronger than that of UA.…”
