Urocortin-1 Infusion in Normal Humans

Abstract: Urocortin-1 (Ucn-1), a member of the corticotropin-releasing factor family, has been shown in animal studies to have effects on the pituitary-adrenal axis, the cardiovascular system, circulating neurohormones, and renal function and to suppress appetite. For the first time in man we have evaluated these effects of infused Ucn-1 as well as actions on plasma ghrelin, a hormone known to increase appetite. We also assessed Ucn-1 pharmacokinetics. Eight healthy male volunteers consuming a diet of constant sodium an… Show more

“…All CRFpeptide family members improve cardiac function via binding to cardiac CRF 2 receptors when administered intravenously. In addition, consistent with CRF-mediated responses, Ucn I has been shown to stimulate ACTH release in healthy rats (63) and humans (12), as well as in sheep (49) and humans (11) with HF via binding to the CRF 1 receptor in the PVN. Although the influence of HF on endogenous Ucn-1 production is unknown, it is possible that Ucn-1 concomitantly with CRF may contribute to the neural-humoral activation characteristic of HF as the CRF 1 and CRF 2 receptor subtypes (11) for Ucn-1 and CRF are expressed in PVN tissue (26).…”

“…Exogenous treatment with somatostatin and its analogs, such as octreotide, as well as infusion of urocortin-1, a potent anorexigenic peptide, suppress plasma ghrelin concentration (17,60,100,177,231). However, administration of leptin does not modify ghrelin levels (36).…”

“…Urocortin 1 radioimmunoassay was performed on plasma samples, as described previously. 31 Sample preparation included steps previously used to extract total CRF (boundϩfree) from human plasma. One milliliter of plasma was mixed with 2 mL methanol and centrifuged.…”
