Urine of patients with early prostate cancer contains lower levels of light chain fragments of inter‐alpha‐trypsin inhibitor and saposin B but increased expression of an inter‐alpha‐trypsin inhibitor heavy chain 4 fragment

Abstract: The present study was aimed at the identification of proteins that are differentially expressed in the urine of patients with prostate cancer (PCa), those with benign prostatic hyperplasia (BPH) and age-matched healthy male control subjects. Using a combination of 2DE and MS/MS, significantly lower expression of urinary saposin B and two different fragments of inter-alpha-trypsin inhibitor light chain (ITIL) was demonstrated in the PCa patients compared to the controls. However, only one of the ITIL fragments … Show more

“…Elevated levels of ITIH4-derived peptides have been observed in serum of breast cancer patients [54]. Increased levels of ITIH4 fragments have also been found in urine of early prostate cancer patients [55]. Reduced levels of ITIH4-derived peptides have also been observed in urine of ovarian cancer patients [56].…”

“…There are a few reports that proposed this protein as an OC marker [37, 42]. Changes in the levels of ITIH4-derived peptides were also observed in urine of early prostate cancer patients [43] and in the serum of breast cancer patients [44] and gastric adenocarcinoma patients [41]. A correlation was also suggested between different fragmentation of ITIH4 and disease conditions [45].…”

“…Protein AMBP, secreted by kidneys and gallbladder, has also been found in studies analyzing blood [77,81] and urine [97]. Other acute response proteins such as SERPINA1, TTR and ITIH4 have also been suggested as candidate biomarkers in serum [78] and urine [97,98].…”

“…Nine proteins with differential abundances were acute phase response proteins and the expression pattern of 4 differed from the defined expression in the canonical pathway. Worth mentioning is that among these proteins was inter-alpha-trypsin inhibitor (ITIH4) which already was reported to be up-regulated in the urine of PCa patients in the study of Jayapalan et al, [98]. The urine levels of TF, AMPB and HP were validated and the expression level was confirmed by immunoturbidimetry.…”
