Nosocomial infections are ranked as the second-largest infections. E. coli is the primary causative agent of this dreadful infection. To develop effective antibiotic substances against hospitalacquired infections, herein a novel carbon monoxide tagged Ru(II) complex of N-o-hydroxyacetophenone-ethylenediamine-N -o-vanillin is reported as an effective antimicrobial agent of its form. In addition to molecular structure elucidation, an explanation has been made about the related charge topography concerning the biological effect. The formulation of the complex based on hyphenated experimentaltheoretical evaluation suggests an octahedral structure. Density functional study was carried out confined to the formalism as B3LYP functional/LanL2DZ basis set for Ru atom and 6-31 g(d,p) for the rest of the atoms. The stable form of geometry and other electron density plots are the prominent inclusions discussed in this work. The primary characterization techniques that have been used for the formulation include elemental analysis, mass spectrometry, nuclear magnetic resonance (C and H), UV-Vis, and FT-IR. The experimentally observed data was validated with theoretical/computational outputs. Both the experimental and theoretical results sound in excellent agreement when compared at each step of the characterization. A suitable trans-octahedral geometry has been suggested for the complex. From the antimicrobial actions against nosocomial E. coli, an MIC value of 30 μM was obtained for the selected compound and hence can be assumed as a standard novel antibiotic substance against nosocomial infections. The respective microbial study has been discussed, keeping in view the microbial growth study, the qualitative and quantitative analysis to add fascination to the work being reported.

Two tetradentate dibasic chelating Schiff base iron (III) chelates were prepared from the reaction of 2,2′-((1E,1′E)-(1,2-phenylenebis(azanylylidene))bis(methanylylidene))bis(4-bromophenol) (PDBS) and 2,2′-((1E,1′E)-((4-chloro-1,2-phenylene)bis(azanylylidene))-bis(methanylylidene))bis(4-bromophenol) (CPBS) with Fe3+ ions. The prepared complexes were fully characterized with spectral and physicochemical tools such as IR, NMR, CHN analysis, TGA, UV-visible spectra, and magnetic moment measurements. Moreover, geometry optimizations for the synthesized ligands and complexes were conducted using the Gaussian09 program through the DFT approach, to find the best structures and key parameters. The prepared compounds were tested as antimicrobial agents against selected strains of bacteria and fungi. The results suggests that the CPBSFe complex has the highest activity, which is close to the reference. An MTT assay was used to screen the newly synthesized compounds against a variety of cell lines, including colon cancer cells, hepatic cellular carcinoma cells, and breast carcinoma cells. The results are expressed by IC50 value, in which the 48 µg/mL value of the CPBSFe complex indicates its success as a potential anticancer agent. The antioxidant behavior of the two imine chelates was studied by DPPH assay. All the tested imine complexes show potent antioxidant activity compared to the standard Vitamin C. Furthermore, the in vitro assay and the mechanism of binding and interaction efficiency of the tested samples with the receptor of COVID-19 core protease viral protein (PDB ID: 6lu7) and the receptor of Gram-negative bacteria (Escherichia coli, PDB ID: 1fj4) were investigated using molecular docking experiments.

Nitric oxide (NO) along with Carbon monoxide (CO) and Hydrogen Sulphide (H2S) are biologically significant gaseous molecules generally called as “gasotransmitters”. At a concentration higher or lower than optimum value may result in toxicity or malfunctioning of mammalian tissues. Soon after the acknowledgment of NO as multifunctional bio-signalling molecule in 1987, many interesting implications of this field emerged out. Meanwhile, several studies have proven the NO-biosynthetic pathway responsible for normal functioning of eye. High intraocular pressure (IOP) has been suggested as the main risk factor in this context and collaborative approach with nitric oxide releasers is said to control IOP and hence the relation with glaucoma. Similar miracles were reflected from several other naturally produced gaseous molecules,viz., CO and H2S after year 1990. The biological roles of both these molecules are now widely accepted and in the current era investigations focused mainly with development of efficient CO and H2S releasing compounds. CO and H2S donors are also said to help in normalising IOP like NO. Therefore the trio-gasotransmitters have collective relation with the ophthalmic homeostasis in association with nervous control. On one hand, the antimicrobial efficiency of these three molecules is widely known and on the other hand, their collaborative key-role in ocular nerve functioning makes it remarkable to state here that their donors are supposed to act as a shield for both the infectious as well as the non-infectious eye defects.
