Urinary metabolite profiling identifies novel colonic metabolites and conjugates of phenolics in healthy volunteers

Abstract: We have positively identified metabolites and conjugates, some novel, in the urine of healthy volunteers after intake of multiple phenolics from a mixed puree from berry fruits, with each being excreted at specific and signature times. Some of these compounds could potentially be used as biomarkers of fruit intake. The possible biological activities of these colonic metabolites require further assessment.

“…However, the determination of this inter-individual variability is often underreported and most works present the data as average or mean and use the standard error of the mean as a dispersion parameter. Data concerning inter-individual variability after (poly)phenol intake has somewhat been buried in the published scientific literature in the form of plasma concentration ranges [31,33] or box-and-plot whisker graphs [39,40], which give a more accurate picture of the actual variation in a dataset but very few papers have described the CV for each kinetic parameter after (poly)phenol intake. We recently reported that the CV of the AUC of the sum of epicatechin metabolites was 38%, which was comparable to the CV of acetaminophen, consumed by the same individuals [35].…”

“…2 and 5). Human studies in vivo and gut microbiota-fermentation studies have demonstrated that at least some of these C 6 -C 1 and possibly C 6 -C 2 phenolic acids can be decarboxylated yielding the corresponding phenols (C 6 ) or methyl-phenols (C 6 -C 1 ) [84,[86][87][88][89][90], respectively. Phloroglucinol can also be formed from chalcones and dihydrochalcones [91,92], and at least certain rumen microorganisms can reduce this to non-aromatic dihydrophloroglucinol [93,94].…”

“…A confirmação da identificação dos metabolitos foi efetuada por LC-MS/MS através da comparação com compostos padrão sintetizados química ou enzimaticamente. Vários metabolitos foram identificados nas amostras de urina, maioritariamente na forma conjugada com grupos metilo, ácido glucurónico e sulfato, sendo que alguns foram identificados em amostras humanas pela primeira vez (PIMPÃO et al, 2014).…”
