Urinary biomarkers of oxidative/nitrosative stress in healthy smokers

Abstract: Urinary 8-OHdG, 15-F(2t)-IsoP and AGEs may represent a non-invasive quantitative index of oxidant stress in healthy smokers, being AGEs a possible indicator of tobacco toxin exposure. The increased oxidative stress in healthy smokers observed may be generated because of an excessive production of reactive oxygen species and not by exhaustion of antioxidant defenses.

“…The elevated 8-OHdG levels in self-reported smokers reflect that smoking leads the extra DNA damage. These results are consistent with previous studies [22,23,32]. Campos et al [22] found that oxidative stress from cigarette did not appear in former smokers.…”

“…On the other hand, Asami et al [21] found the level of 8-OHdG in lung tissues was 1.43-fold higher in smokers than in non-smokers. Campose et al [22] have also found recently higher urinary 8-OHdG levels in healthy cigarette smokers than in non-smokers (10.7 ng/mg creatinine vs. 8.3 ng/mg creatinine). Increased excretion rate of urinary 8-OHdG also has been associated with cigarette smoking, even in lung cancer patients [23].…”

“…Elevated levels of isoprostane 8,12-iso-iPF2α (iPF2α), a specific marker of lipid peroxidation, have been demonstrated in living adults with DS [13]. Recently, evaluation of some biomarkers of oxidative/nitrosative stress in the urine from adults compared with adolescent DS subjects demonstrated that advanced glycation end products, dityrosine, H 2 O 2 and nitrite/nitrate could be considered as OS biomarkers in DS in contrast to other markers, such as 8-OHG, 15-F(2t)-IsoP and TBARS, which gave contrasting results [14]. However, additional studies on large populations are needed to confirm the reproducibility of these results.…”

“…Data regarding cigarette smoking is controversial. While some studies showed an association between cigarette smoking and 8-OHdG levels (Campos et al, 2011;Chiang et al, 2012), others failed to show such an association (Feng et al, 2006). Previous studies revealed higher levels of 8-OHdG in the lung, liver, and heart of mice exposed to cigarette smoke (Howard et al, 1998;Thaiparambil et al, 2007).…”
