“…MATERIALS AND METHODS Purified synaptosomes (17) from adult rat cerebrum resuspended in 0.32 M glucose were diluted 1:10 in a Krebs-Ringer medium [128 mM NaCl/5 mM KC1/2.7 mM CaCl2/1.2 mM MgSO4/1 mM Na2HPO4 20 mM N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (Hepes) buffered at pH 7.35], equilibrated for 10 min at 370, and then labeled for another 10 min with 0.5 uM [3H]GABA (New England Nuclear Corp., specific activity 30 Ci/mmol). After the labeling period, a tissue/medium ratio of radioactivity of 70-90 was obtained.…”
