Uptake and metabolism of retinol in cultured Sertoli cells: evidence for a kinetic model

Abstract: When cultured Sertoli cells derived from 20-day-old weanling rats were supplied [3H]retinol bound to serum retinol binding protein-transthyretin complex, [3H]retinol was rapidly incorporated and [3H]retinyl esters were synthesized. Within 28 h after administration, 83% of the labeled retinoids were accounted for as retinyl esters (64% as retinyl palmitate). Sertoli cells derived from vitamin A deficient rats and supplied [3H]retinol in culture under identical conditions likewise incorporated [3H]retinol and sy… Show more

“…This is consistent with peroxisome proliferators possibly countering the effects of endogenous retinoids in primary Sertoli cells. Sertoli cells are known to store retinoids [40]. Considered together, these results clearly suggest that peroxisome proliferators disrupt the retinoid signaling pathway also in primary Sertoli cells.…”

“…In many ways, the liver and Sertoli cells appear to function similarly in the uptake of retinol and the maintenance of large pools of this retinoid (35,43). However, it seems that spermatids and testicular and epididymal sperm also store retinoids (44).…”

“…Based on measures made in rat liver and testis that show CRBP I is present at concentrations in excess of those of retinol, it is believed that all retinol present within these tissues is bound to CRBP I (HARRISON et al 1987;BISHOP and GRISWOLD 1987). It is generally accepted that CRBP I plays an important role in retinol metabolism (ONG et al 1994a).…”
