Upregulation of prolylcarboxypeptidase (PRCP) in lipopolysaccharide (LPS) treated endothelium promotes inflammation

Abstract: BackgroundProlylcarboxypeptidase (Prcp) gene, along with altered PRCP and kallikrein levels, have been implicated in inflammation pathogenesis. PRCP regulates angiotensin 1–7 (Ang 1–7) – and bradykinin (BK) – stimulated nitric oxide production in endothelial cells. The mechanism through which kallikrein expression is altered during infection is not fully understood. Investigations were performed to determine the association between PRCP and kallikrein levels as a function of the upregulation of PRCP expression… Show more

“…Moreover, inhibition of tyrosine phosphatase with sodium orthovanadate abolished AT 2 R-stimulated plasma prekallikrein activation and bradykinin release. Because plasma prekallikrein activation is an indirect measure of PRCP activity (18,25) and PRCP is known to have several tyrosine phosphorylation sites, our data suggest that AT 2 R-stimulated PRCPdependent plasma prekallikrein activation is mediated at least in part via SHP-1.…”

“…For kallikrein activity, we followed the methods established by Ngo et al (18) and Shariat-Madar et al (24). Cells seeded in 96-well plates were washed three times with HEPES-NaHCO 3 buffer containing 137 mM NaCl, 3 mM KCl, 10 M CaCl2, 1 mM MgCl2, 12 mM NaHCO3, 5.5 mM dextrose, 0.1% gelatin, and 14.7 mM HEPES (pH 7.1).…”
